Background and Objective There are many changes of energy metabolism in hypertrophy myocardium, such as changes of metabolism material,energy-producing structural,systolic proteinduring the process of compensatory- uncompensatory. Decrease of energy reserve, increase of energy expend,accumulation of metabolism aggravated compensatory overload upon hypertrophy myocardial , shortage of energy production may play a major role in energy metabolism during development of Hypertrophy myocardial. Astragalus (Huangqi) , one of the best Supplementing qi Chinese Herb, was first recorded in "Shennong Chinese Herb Classic". Research proved that Huangqi prevented myocardium and hypertrophy myocardium from ischemia-reperfusion injuring,oxygen-derived injuring by protecting the structure and function of mitochondria,increasing activity of enzyme,reducing the depletion of ATP, accommodating energy metabolism. Huangqi injection could strength myocardial heartbeat,improve energy metabolism,protect myocardial ultrastructure,increase the activity of SOD and increase the ATP level of myocardial. Huangqi injection was used to cure the diseases such as endomyocarditis,coronary heart disease,heart failure. To study the effects and mechanisms of Huangqi on the cardiac function and energy metabolism of Hypertrophy myocardial, the experiment use advanced appearance and investigate the effect of Huangqi injection on change of hypertrophy myocardial activity induced by angiotentin â…¡,enzyme co relational energy metabolism, change of mitochondrial membrane potential(MMP) ,which referred the reference for Traditional Chinese Medicine preventing and curing ventricular hypertrophy and heart failure. Material and Methods According to Harary's method, we use cultured neonatal rat cardiomyocytes to explore. There are four group in this experiment: control group, model group, lorsartan group and therapy group. The models of the myocardial hypertrophy induced by angiotensin â…¡ were established by observing the content of the total cellular protein. In addition to the above parameters, the studies were also done to understand the effects of Huangqi injects, in 6 concentrations of 10-3g/L,10-4g/L,10-5g/L,10-6g/L,10-7g/L,10-8g/L , on the myocardial hypertrophy induced by angiotensin â…¡ in the cultured neonatal rat cardiomyocytes in vitro. Then the studies investigated the effect of Huangqi injection on the activity of SDH,LDH, the MMP change of hypertrophy cardiomyocytes induced by angiotentin â…¡ by laser confocal scanning microscopy(LCSM). Results Total protein of Angâ…¡ group were much high than those in Control group (P<0.01). The activity of hypertrophy myocardial induced by Angâ…¡ increased at time dots 12h and 24h but decreased at time dots at 48h ,72h ,96h. The activity of hypertrophy myocardial of Huangqi 10-3g/L group,Huangqi 10-4g/L group was lower than that of Lorsartan group,Control group(P<0.01); the activity of hypertrophy myocardial of Huangqi 10-5g/L group, Huangqi 10-6g/L group,Huangqi 10-7g/L group were lower than that of Angâ…¡ group(P<0.01),and at the same time there was no difference between that of Huangqi 10-5g/L group,Huangqi 10-6g/L group,Huangqi 10-7g/L group and that of Lorsartan group,Control group(P>0.05). Huangqi 10-6g/L group was chosen . The activity of SDH of Angâ…¡ group was much higher than that of Control group(P<0.01); the activity of SDH of Lorsartan group,Huangqi group, Lorsartan+Huangqi group were lower than that of Angâ…¡ group(P<0.05),but there was no difference between Lorsartan group, Huangqi 10-6g/L group, Lorsartan+Huangqi group and control group(P>0.05).
