Chinese Medicine Polysaccharide Anti-tumor Effect And Its Mechanism In The Signal Transduction Pathway

Objective :To study the antitumor functioning and probe its under lying mechanism of plant polysaccharide in cellular signalling pathway of PIP3/Ca2+-DAG/PKC,and confirm the new target of Chinese medicinal herbs from the three ways of calcium ion, protain kinase c and cell apoptosis, and explore their relations.Methods: we treated mice, which mouse S180 sarcoma cells had been inplanted in subcutancous oxter-region with three kinds of herbs. The tumor inhibitory rate and the expression of protain kinase c were tested in animal tumor model with experimental treatment by immunistological method. we treated mouse S180 sarcoma cells with three kinds of herbs. The cell inhibitory rate was tested through MTT ;After cultured for 1 and 8h, The cell apoptotic rate of mice sarcoma S-180 cell line was observed by flow cytometry; After cultured for 1 and 8h, The chang of intra-cellular calcium ions of mice sarcoma S-180 cell line was measured by immunofluorescence method.According to the results,we probe its under lying mechanism.Results: When the different doses were used , the tumor inhibitory rate of the three herbs groups PB (polyporusum bellatus) PPS(pachyman polysaccharides) APS(astragalus polysaccharides) were 63%, 54% ,47%, and 30%,36%, 42%,and 37%, 67%, 55% respectively ; and the expression of protain kinase c in the groups of PB (polyporusum bellatus) were up-regulated compared with the control group (p<0.05),others have no significant difference(p>0.05); calcium ion and cell apoptosis of mice sarcoma S-180 cell line was induced by PB ,PPS and APS in dose-and time-dependent manners.After respectedly treated with100. 250. 500 ug/ml of Plant polysaccharide for 1 and 8h. calcium ion and cell apoptotic rate of mice sarcoma S-180 cell line were on the rise with time and dose, the change of calcium ion and apoptotic rate of experiment groups have significant difference compared with control group(p<0.05). Conclusion: The three drugs have inhibitory effect on S180 sarcoma . Plant polysaccharide may induce cell apoptosis of S-180 cell line in early phase and induce necrocytosis in late phase,inbihit poliferation of mice sarcoma S-180 cell; and the expression of protain kinase c was up-regulated;and intra-cellular calcium ions have obvious increase in different periods .
