Objective: To evaluate the feasibility of using abulmin nanoparticles asp53 gene carries.Methods: Abulmin nanaparticles (ANP) were prepared by coacervationprocess. P53 were absorbed electrostatically on p53 on albuminnanoparticles. The potential of absorbing DNA and protection DNA wasanalyzed by electrophoresis. P53-ANP, p53-liposome and empty DNAwith the same quantity were transfected to HepG2. P53 gene expressionwas analyzed by PR-PCR at mRNA level and Western-Blot at proteinlevel.Results:1. electronical microscope found: Albumin nanoparticles were average dispersed particles, with sound average diameters of 98nm.2. electrophoresis showed: albumin nanoparticles can effectively absorb and protect p53 genes at acid PH.3. PT-PCR and Western-Blot indicated: albumin nanoparticles could transfer p53 gene into HepG2 cells and expressed in vitro.4. Growth curve showed: p53-ANP more effectively inhibitor the growth of HepG2 cells compared with that of liposome group (p<0.05). Conclusion: ANP were prepared with sound diameter, and adsorpted p53 efficiently. The p53-ANP can transfect HepG-2 and inhibitor its growth. Thus, ANP are elucidated the potential as gene carrier.
