Objective: To research the effect of the gene expression profile in "MSG- liver regeneration-rat" by Zuogui Pill, and to study the molecular mechanism of liver regeneration disturbance in "MSG-liver regeneration-rat" and Zuogui Pill adjusted its liver regeneration.Methods: Copying "MSG- liver regeneration- rat" model, treated group was treated by Zuogui Pill (5g/kg), model group and control group were treated by distilled water with equal volumes, then, 2 weeks later, all the rats were operated to excise the left and middle lobe of liver (about 68% of all the liver). The treatment after operation was same as before. Rats were executed at 5th day after operation, and the regenerational liver tissues were froze by liquid nitrogen. The gene chip of gene expression profile concluded 1176 genes that were correlated to cell differentiation and multiplication were used to detect the specimens. Extracted mRNA of the regenerational liver tissue to make cDNA probe by RT-PCR and adding 33P to the probe. The cDNA probe that labelling by 33P hybridizated with the gene chip to examine the change about gene expression profile of the regenerational liver tissues. The data was treated with Microsoft Access and Excel to filter the differently expressed genes, which ratio of hybridized signal between the swatchs had double diversity.Results: In the examined 1176 genes, there're 256 differently expressed genes that model group contrasted to control group, and 40 genes were higher expressed, 216 genes were lower expressed. In the 256 differently expressed genes, there're 10 genes that associated with cell receptors, 15 genes that associated with transcription, 9 genes that associated with cell adhesion receptor, 98 genes that associated with metabolism, 9 genes that associated with post-translational modification/protein folding, 11 genes that associated with protein turnover. There're 292 differently expressed genes that treated group contrasted to model group, and 20 genes werehigher expressed, 272 genes were lower expressed. In the 292 differently expressed genes, there're 28 genes that associated with transcription, 11 genes that associated cell adhesion receptor, 35 genes that associated with metabolism, 2 genes that associated with oncogene and tumor suppressors, 5 genes that associated with post-translational modification/protein folding, 2 genes that associated with DNA binding and chromatin proteins, 4 genes that associated with RNA processing turnover and transport, 32 genes that associated with protein turnover, 14 genes that associated with cytoskeleton/motility proteins, 2 genes that associated with DNA synthesis recombination and repair. Zuogui Pill can aim directly at adjusted 12 genes' expression. Furthermore, there're 5 genes that are upper expressed when model group contrasted with control group, and lower expressed when treated group contrasted with model group. GenBank ID of the 5 genes are AF049344, D83948, L08812, U53883, U55938. Gene expression product of AF049344, U53883, U55938 belonged to protein modification enzymes. Gene expression product of D83948 was chromatin protein. Gene expression product of L08812 belonged to basic transcription factors. There're 7 genes that are lower expressed when model group contrasted with control group, and upper expressed when treated group contrasted with model group. GenBank ID of the 7 genes are Dl 1444, M81786, M86758, U24282 U53505, U89743, X84004. Gene expression product of M86758 correlated with metabolism of cofactors, vitamins, and related substances. Gene expression product of U24282, U53505 were other metabolism enzymes. Gene expression product of M81786 was matrix adhesion receptor. Gene expression product of D11444 belonged to growth factor, cytokine, and chemokine. Gene expression product of X84004 belonged to intracellular protein phosphatases.Conclusions: The gene expression profile of the regenerational liver tissue in "MSG- liver regeneration- rat" was marked different contrasted with normal partial hepatectomy-liver regeneration-rat.
