Objective: The selective accumulation of endogenous PpIX in rats colon tissue following systemic administration of 5-ALA with the dose of 25mg/kg wt and interval time of 150min has been examined. The correlation of the PpIX concentration distribution in blood and tissue with pathological change has been studied.Materials and Methods: Multifocal colon cancers were induced by 25mg/kg.wt injections of 1,2-DMH in SD rats, the second era of induced rats and normal SD rats. Chemical extraction assays of the PpIX content in tissue and blood were carried out at 150 min after trail intravenous injections of 5-ALA at a dose of 25mg/kg.wt with fluorescence spectrophotometer.Results: A significant difference of PpIX content was found in tissue and blood, with a mean of 2363.44 +241.25ng/ml, 3357.39 + 369.11 ng/ml, 4939.55 + 692.68 ng/ml in blood and 622.97 + 61.85 ng/g, 763.098 + 83.93 ng/g , 844.16 + 87.56ng/g in tissue respectively corresponding to advanced cancer, early cancer and normal rats.Conclusions: Systemic administration of low dose 5-ALA can induce effective tissue sensitization with PpIX and appears to be a promising technique for PDD.
