Objective: The manufacture of Galactosed human serum Albumin Magnetic Adriamycin Nano-Particle (GAMANP) under the optimium conditions.and through the one-off jugular vein injection on home rabbits with different two drugs, to check-up whether the pharmacokinetic of GAMANP is different to the Adriamycin or not, and find out the pharmacokinetic characteristics of the new drug. Methods: Use reductive lactosam to synthesize galactosylated albumin, then use heating curdle to synthesize GAMANP, to measure the diamete of the nanoparticle under the Laser Granularity Analyse Apparatus.and measure the enveloping rate and the loading quantity under the ultraviolet prismatic photometer. The twelve home rabbits were divided into two groups in random, six each. Every rabbit was put a pipe in its jugular vein, then injected drugs and exsanguinated. One group was given Adriamycin 3mg/kg to every rabbit, the other group was given GAMANP 61.49mg/kg to every rabbit, exsanguinated 1ml in each time, then separated serum and saved in low temperature,distill the Adriamycin with chloroform-carbinol, put it under the High Performance Liquid Chromatography (HPLC) to examine the special hromatogram apex height. Finally, account the concentration of the Adriamycin in serum according to the standard curve of the Adriamycin, Then analyse the test date with "SPSS" software and pharmacokinetic parameter in "3P87"and"MATLAB "software. Results: a-Lactose and albumin reacted about 60 hours, the galactosylated rate of human albumin is 56. 24 ug/mg,the average diameter of GAMANP is 232nm, loading 47.5+4.57ug adriamycin per 1 milligram, and the percent of adriamycin envelopment is 94.34+3.12%. The pharmacokinetic of Adriamycin and GAMANP in home rabbits were according to the three chamber models.Compared to Adriamycin, the half life of GAMANP was prolonged about from 1.9 to 3.2 tunes; and the clearness of new drug from blood was 0.5369 times to Adriamycin, and the Area ofUnder the Curve(AUC) was 1.3697 times to Adriamycin.
