ObjectiveWe established the rats in diabetic myocardiopathy model.Observing the effect of the Fufangkuqiaomai (FFKQ ) on the blood glucose, body weigh,index of left ventricle (left wentricle weight/body weight), blood TC, HDL-CH, lipoprotein LDL-CH, TG, ultrastructure of myocardial cells,the expression of GLUT4 and the staining of HE and PAS in myocardial tissue in diabetic rats induced by injecting STZ into abdomen.This experiment was to investigate the mechanism of the protective effect on the early diabetic myocardiopathy of FFKQ and provide an experimental basis for further investigating and clinical use. MethodsThe diabetic rats were induced by injecting STZ into their abdomen successfully,and they were stochastic divided into four groups: FFKQ high dose group ( high dose group ) , FFKQ low dose group (low dose group ) , xiaokewan group, model control;the other normal rats were the normal control group ( normal group.) The respective dosage of drugs was given to the treated groups and the same dose of sodium chloride injection was given to the normal and model series by stomach rearing once per day for eight weeks.After therapy,the body weight, the blood glucose,index of left ventricle,blood TG,TCH,HDL-CH,LDL-CH,ultra-structure of myocardial cell,expression of GLUT4 and the staining of HE and PAS in myocardial tissue of each group were tested.All the results were analyzed by SPSS 11.0 for windows. Result1 Body weight There were no significant differences among all groups before treatment,P > 0.05.After treatment,the body weight of model group kept on decreasing,while that of normal group kept on increasing,which indicidated there was very significant difference between them,P < 0.01.The body weights of the high dose group,low dose group,xiaokewan group were distinctly higher than that of the model control group,P < 0.01.The weight decreasing degree of high dose group was lower than that of the low dose group and xiaokewan group,which indicated there was no significant difference among them,P > 0.05.2 Blood glucose There were no significant differences among model group and all therapeutic groups in blood glucose before treatment,P > 0.05,but the levels of blood glucose in all these groups were higher than that of the normal group, P<0.01;after treatment,the levels of blood glucose in all therapeutic groups were lower than that in model group and before treatment,P < 0.01,and the high dose group had better effect on lowering the level of blood glucose.3 Serum lipid (1)triglyceride ( TG ) : TG in all therapeutic groups was distinctly lower than that in model group,P < 0.01,and the high dose group had the best effect among them.(2)low density lipoprotein cholesterol ( LDL-CH ) : LDL-CH in all therapeutic groups was distinctly lower than that in model group, P < 0.01,and thehigh dose group had the best effect among them.(3)total cholesterin ( TC ) : TC in high dose group was distinctly lower than that in model group, P<0.01.(4)high density lipoprotein cholesterol (HDL-CH ) : HDL-CH in all therapeutic groups was distinctly higher than that in model group, P < 0.01,and the high dose group had the best effect among them.4 Index of left ventricle Compared with the model series,the high dosage and xiaokewan group could markedly decrease the index of left ventricle, P < 0.01.There were not distinct difference between the effect of the high dosage and xiaokewan group.5 the expression of GLUT4 Compared with the model series,FFKQ could obviously increased or promoted the expression of GLUT4 in the myocardial.The best group were the high dosage group and xiaokewan group.6 the staining of HE and PAS The HE's staining showed that the model group rat's myocardial fiber soluted, degenerated and necrosed partly.The PAS's staining showed thatthere were large quantity of glycogen that deposited in the model group rat's myocardial tissue (show the carmine or solferino ) .Each other series could prevent myocardial tissue and fiber from these changes.7...
