Owned Red Particles In Ovarian Granulosa Cells Produce Estrogen Effects

Objective: To study the effects of Zichongkeli and its components on the estrogen production in cultured mouse ovarian granular cells, which helps to explain the mechanism that Zichongkeli is able to stimulate follicular development.Method: Mouse ovarian granular cells were cultured in RPMI 1640 medium containing 10% calf serum. Aftet 24 hours, different concentrations of testosterone propionate were added to the medium and ovarian granular cells were cultured for 24 and 48 hours. Under the condition that testosterone propionate was added to the medium,Zichongkeli and its components and serum containing Zichongkeli were added to ovarian granular cells which were cultured for 48 hours. IGF-1,FSH and Nujindan were taken as control group. Estradiol which was transfered from testosterone in the medium were measured by Enzyme immunoassay.Results: Estradiol production in cultured mouse ovarian granular cells was related to the concentration of testosterone propionate and its cultured time.Zichongkeli and Nujindan could promote the quantity of Estradiol transfered from testosterone in the medium,but less than the level that stimulated by IGF-1. Compared with the blank group, Shudi and Tusizi had an action of promoting estradiol transfered from testosterone in the medium,while Chuanxiong's had no effect on mouse ovarian granular cells. The group which tonifys kidney had no action of promoting Estradiol transfered from testosterone in the medium, while the group which promotes blood circulation and dissipates blood stasis had an action of suppressing on estradiol transferred from testosterone in the medium.Serum containing Zichongkeli had obvious effect on promoting estradiol transfered from testosterone in the medium.Conclusion: To promote estradiol production in cultured mouse ovarian granular cells is one of mechanisms of Zichongkeli in stimulating follicular development.
