Objetive:To obtain a complete X gene sequence of the hepatitis B virus, provide the gene material for "the development of detecting kit of x antigen in hepatitis B virus", and the studies of biologic functions of HBV X gene and X gene-encoded protein.Methods:HBV X gene was amplificated by the Polymerase chain reaction(PCR) from sera of the Hepatitis B patients. Using the clone system of the plasmid pUC 18 and Escherichia coli JM109 to orientation cloning. And determinated this recombinants by restriction enzyme zymogram Analysis, PCR and DNA sequencing.Results:We acquired A series of reconstructed plasmid PUC18-X that carrying Integra X gene sequence.Conclusion: It is proved that the recombinated plasmid PUC18-X earring HBV X gene sequence. This experment will be a foundation for the following studies of HBx.
