Pedigree is one of the most general population structure in the genetic linkage analysis between human single genetic inheritance disease locus and genetic marker locus. In pedigree genetic linkage analysis, the identity of marker alleles carried by any two diploid individuals is generally regarded as their genetic correlation measure about this marker locus, for example, most genetic linkage analysis methods are based on affected sib pair(ASP); the transmission/disequilibrium test(TDT) is based on parents and their affected child; the IBD-APM genetic linkage analysis method is based on the pedigree of an affected individual. But, it is difficult to tell the identity of two alleles. This paper is aimed at constructing genetic linkage analysis on the genetic correlation measure of marker genotype of any relative pair with some genetic relation; using the Pearson x2 statistic to test the genetic linkage between disease locus and marker locus. Moreover, it analyzes the power of Pearson x2 test by simulation.Chapter Two is the foundation of full paper, and it constructs a model about the joint distribution of the genotype on two linkage loci of any relative pair with some genetic relation. The model in Chapter Two is an extension of the joint distribution model on single locus of any relative pair with some genetic relation, which was set up by C.C. Li in 1954. |
