The brain is the neurophysiological basis for the functioning of the brain. Learning and memory is one of the major functions of the brain. Reports from recent years have shown that some traditional Chinese medicines are effective in improvement of function of the brain. NGFD, developed by our department had been proven to work effectively in improvement of function of the brain. But the mechanism is still need to be explored. It is widely accepted that taurine plays an important role on neuroprotection. However, the clinical application of taurine is more or less limited because of the low permeability to blood-brain barrier(BBB) which results from its inferior lipophibic property .Therefore, (1)we studied the effect of NGFD on learning and memory in both developing and aged brain exploring its possible mechanism, (2)In order to provide some experimental data for clinical neuroprotection application, the permeability of esterized taurine through BBB was studied.In the present study, praxiology training, immunocytochemistry(ICC) staining~. flowcytometry (FCM) TDT-mediated ddUTP nick end labeling (TU7NEL) and amine acid assaying etc. techniques were used. The experiment was designed as 3 parts. Part I Experimental study on the effect of NGFD on the function of developing rat brain The effect of NGFD on developing rats capacity of learning and memory as well as possible mechanism were studied. 0.5 month old rats were randomly divided into control group(normal feed) and experimental group(2m1 NGFD/day). After feeding for 2 monthes , animals were trained on active avoidance reactive(AAR) and passive avoidance reactive(PAR), then sacrificed for examination of changes of the synaptosomeN synaptophysin(SY) by ICC. Result:increased AAR acquisition and decreased AAR extinction were observed. The step through latency(STL) of PAR was significantly prolonged. The number for synaptosome and the immunoreactive intensity for SY were increased obviously. It suggested NGFD was beneficial to the improvement of the function of developing rat brain. Part II Experimental study on the effect of NGFD on aged rat brain To evaluate the effect of NGFD on aged brain of the rats and its possible mechanism. Rats were first divided into A and B group according to the results of active avoidance reaction (AAR) test. Then each group was randomly divided into control and NGFD-treated experimental groups. Animals were feeded with NGFD for 2 months (5ml/day) and then sacrificed for examination of changes of synaptosome~ SY~ Tau protein and apoptosis in cerebrum as well as behavior ( AAR, PAR). Results: Increased AAR acquisition and decreased AAR extinction were observed, and STL of PAR was significantly prolonged . The number for synaptosomes and the immunoreactive intensity for synaptophysin were increased obviously, while Tau protein-immunoreactivity and apoptotic cells were decreased in cerebrum. It suggested NGFD have the effect in improving brain function and putting off the aging of rat brain.
