Purpose:To explore the angiogenic mechanism of eye acupuncture treating cerebral ischemia reperfusion through established the cerebral ischemia reperfusion injury model in rats, the expressions of vascular endothelial growth factor (VEGF),Angiopoietin-1 (Ang-1) and Endostatin in the brain tissue of the rat were detected with immunohistochemisty SABC.Material and method:40 SPF Healthy male Wistar rats, 300±20g in weight, were randomly divided into 4 groups:shame-operated group, model group,body acupuncture group and eye acupuncture group. After the adaptive feeding one week, middle cerebral artery occlusion model was established by improved modified suture method,which was assessed with the Zea-Ionga signs of neurological defect standards after cerebral ischemia reperfusion(CIR). The rats of score reaching level 1,2 and 3 were divided into in the model group, body acupuncture group and eye-acupuncture group. After CIR,the rats in the sham-operated group and model group were not given any treatment, and the rats in the eye acupuncture group were treated with the 25-minute eye-acupuncture treatment in the areas of Shang-jiao, Xiao-jiao, Liver and kidney every 12 hours during animal survival, while the rats in the body-acupuncture group were acupunctured the Quchi Acupoint with the same time. Specimen collection:All rats in the four groups were anaesthetized with Urethane (0.35mL/100g) after successfully establishment of CIR model 24 hours later,then the rapid perfusion of the ascending aorta was carried out in order to take the brain tissue reserved. Immunohistochemisty SABC method was used to investigate the expressions of VEGF,Ang-1 and Endostatin. All data were analyzed with single factor analysis of variance among groups in SPSS10.0 software statistics. Results:1. Effect of eye acupuncture on the rat behavior after CIRIn accordance with the Zea-Ionga signs of neurological defect assessed after cerebral ischemia reperfusion.Compared with those in the sham-operated group,there had the signs of neurological defect in the other three group and TTC Staining displayed the model was successfully established;Compared with those in the model group, the signs of neurological defect decreased and the symptoms alleviated in the body acupuncture group and eye acupuncture group after the acupuncture,which revealed the acupuncture can improved the signs of neurological defect;Compared with those in the body acupuncture group,the signs of neurological defect in the eye-acupuncture group was lower,and this revealed the eye-acupuncture was more effective than the body-acupuncture in the early stage of cerebral ischemia reperfusion。2. Effect of eye-acupuncture on expression of the VEGF in the brain tissue of the rat after cerebral ischemia reperfusionCompared with those in the sham-operated group, the expressions of VEGF in the other three groups were significantly increased(P<0.01); Compared with those in the model group, the expressions increased in the body-acupuncture group(P<0.05)and had significant difference in the eye-acupuncture group(P<0.01);Compared with those in the body-acupuncture group, the expressions of VEGF increased(P<0.05)in the eye-acupuncture group.3. Effect of eye acupuncture on expression of the Ang-1 in the brain tissue of the rat after cerebral ischemia reperfusion. Compared with those in the sham-operated group, the expressions of Ang-1 in theother three groups were significantly increased(P<0.01); Compared with those in the model group, the expressions increased in the body-acupuncture group(P<0.05)and had significant difference in the eye-acupuncture group(P<0.01);Compared with those in the body acupuncture group, in the eye acupuncture group, the expressions of Ang-1 increased(P<0.05).4. Effect of eye-acupuncture on expression of the Endostatin in the brain tissue of the rat after cerebral ischemia reperfusion。 There was almost no expression of Endostatin in the sham-operated group; Compared with those in the model group, the expressions decreased in the body-acupuncture group(P<0.05)and decreased significantly in the eye-acupuncture group; Compared with those in the body-acupuncture group, the expressions decreased in the eye-acupuncture group(P<0.05).Conclusion:1. Eye acupuncture can alleviate the signs of neurological defect in the cerebral ischemia reperfusion model rats, and its early action is better than that of body acupuncture.2. Eye acupuncture can improve VEGF and Ang-1 expressions in ischemic brain area.3.Eye acupuncture can inhibit Endostatin expression in ischemic brain area.
