Objectives:The theory of traditional Chinese medicine (TCM) holds that Qi is the commanderof blood and Qi has the function of promoting the circulation of blood., these instructions Qi has a close ralationgship with blood, exuberant Qi can promot blood circulation in vessel, otherwise, deficiency of Qi cause stagnation of blood, this is that blood stasis due to qi deficiency, the treatment should add some herb of activating blood and resolving stasis, while takeing much herb invigorating qi. Although there is no uniform understanding on the symptome differentiation of heart failture,a number of scholars agure that stagnation of blood is pathological basis of heart failture, hence, therapy of heart failture is mainly activating blood and resolving stasis, while, warming yang for diuresis and nvigorating qi, yet, according to the above TCM theory, a fountainhead of stagnation of blood is Qi, no matter what is deficiency of Qi or Qi stagnation. Relying on the above analysis,this issue study of Yiqihuoxue Medicine(mainly invigorating qi) on the affecting of heart failture treatment,we can reveal Qi-blood theory.myocardial cell apoptosis was found in heart failture rats, Thus, another purpose of this study is to explore TCM in the anti-apoptotic probable mechanism of action.Methods:Based on the research literature, through the pre-experimental trial and error, we established intraperitoneal injection for 3 consecutive days of isoproterenol (5mg/kg/d) method for the construction of the best model of heart failure.the rats were randomly divided into 3 groups:model group, Yiqihuoxue group Tongxinluo Group, together with the normal group a total of four groups.these rats was taken drurs for continuous Several days and would be killed after 42 days, The number of myocardial cell apoptosis is detected by immunohistochemistry in situ apoptosis (TUNEL) of myocardial cell apoptosis, Bal-2 and Bax expression levels is measured by PCR, cytC expression is measured by western blot, Cardiacfunction changes in rats is detected with ultrasound.Results:1 Ultrasonic testing: wo would observed IVSTd, LVDd, LVPWTd, IVSTs, LVDs, LVPWTdsFS and EF after 28 and 42 days for the normal group and including ultrasonic testing of all animals.By statistical analysis, at the point of 28 days, wo found LVSTd, LVPWTd, LVPWTs increased, LVDd, LVDs lower FS. Compared with the normal group, there were significant difference (P<0.05). after 42 days,42 days, wo found LVDd, LVDs increased, IVSTd, IVSTds, LVPWTd, LVPWTs, EF, FS decreased compared with the normal group, there was significant difference (P<0.05), and EF decreased more than 25%; of drug intervention, EF, FS, VSTd, IVSTds, LVPWTd, LVPWTs of Yiqihuoxue and Tongxinluo group was increased significantly, LVDd, LVDs significant ly reduced, compared with the model group there were significantly differences (P<0.05), especially Yiqihuoxue most obvious difference between drug groups.2 Left ventricular index:LV index was increased in model group, compared with the normal group there was a significant difference (P<0.01).Yiqihuoxue drug group and tongxinluo group was lower than model group (P<0.05), Yiqihuoxue drug was the lowest3 Myocardial cell apoptosis:, apoptotic rate of model group was significantly higher than normal group, there was a significant difference (P <0.01)。After drug intervention, myocardial cell apoptosis rate of Yiqihuoxue and Tongxinluo was lower than model group, there was a significant difference (P<0.01); Yiqihuoxue drug is the most significant effective.4 Bal-2, Bax expression: Bal-2 of the model group was significantly lower and Bax was increased, while, Bal-2/Bax also was significantly reduced。There was significant different compared with the normal group (P<0.01); with the model group, Yiqihuoxue and Tongxinlou's Bal-2 were significantly higher, Bax and Bal-2/Bax was significantly increased, there was significant difference (P <0.05), especially Yiqihuoxue was the most effective.5 cytC expression:comparison with the normal group, cytC of model group was significantly higher, there was significant difference (P<0.01); Yiqihuoxue and tongxinluo's cytC was significantly reduced compared with the model there was a significant difference (P<0.01). Yiqihuoxue drug had the strongest effect ive.6 Cardiac function: EF, FS of the model group was lower,, LVEDD, LVESD was increased and I VST, LVPWT was reduced, there was significant difference compared with the normal group (P<<0.05); after the drugs intervention, EF, FS of Yiqihuoxue and Tongxinluo group were increased sign if icant ly,, LVEDD, LVESD were decreased significantly and IVST, LVPWT was significantly increased compared with the model group (P<0.05), especially group Yiqihuoxue was obvious difference.Conclusions:1. Rats were injected isoproterenol for 3 consecutive days, heart failure would be caused, heart function would be lower. Drug intervention can improve heart function of heart failure, Yiqihuoxue drugs was the best.2 Yiqihuoxue can make Bal-2's expression of myocardial cells increased, Bax expression levels decreased and Bal-2 and Bax ratio increased, which can inhibit myocardial cell apoptosis.3 Bax expression levels of the myocardium are closely related in cytC, Bal-2 increased, Bax decreased, Bal-2/Bax increased can inhibit the expression of cytC, otherwise it will promote its expression, which may be the Bal-2, Bax regulation of apoptosis.4 Yiqihuoxue drugs can affect the cytC's level in the myocardial tissue by the regulating of Bal-2, Bax expression, to prevent the downstream apoptotic pathways, thies may be a mechanism that yiqihuoxue drug.treat heart failure. |
