Acupuncture Eae Rat Il-23, Sicam-1 In Rats

Multiple sclerosis (MS) is a chronic inflammatory central nervous system demyelinated disease. It is characterized by affecting young adults, having long course and relapsing-remitting. The pathological hallmark of MS is the sequential development of demyelination. Since the mechanisms of myelin breakdown are still unknown, there is no reliable means for therapy of MS till now. Accordingly, to explore the pathogenesis of MS and to successfully treat MS are required. Experimental autoimmune encephalomyelit is (EAE) is a kind of autoimmune diseases occurring in the Central nervous system (CNS) of sensitive experimental animals. It is a classic animal models of CNS inflammatory demyelina ted disease.This experiment by establishing EAE model of rats, detect ion EAE rats remiss ion IL - 23 and sICAM-1 level after acupuncture, discusses the mechanism of acupuncture in the treatment of EAE, and compare the curative effects of acupuncture treatment with hormone therapy.Methods:49 female Wistar rats were randomly divided into normal group, model group, elec tro-acupunct ure group, and hormone group. Then the female Wistar rats were immuned by a stable water-in-oil emulsion which was made from guinea pig spinal cord homogenate (GPSCH) and equivalent complete Freund's adjuvant (CFA), then injected whooping cough bacili vaccine immune within 72h to prepare the experimental autoimmune encephalomyelitis (EAE) model. Normal group was injected physiological saline on limbs foot pad, then injected physiological saline within 72h. Electro-acupuncture group was acupunctured on baihui, shenshu and zusanli, cupping lateral shenshu and zusanli.Hormone group was given tail intravenous methylprednisolone by sodium succinate and irrigated prednisone acetate macroporous. While normal group and model group were given no treatments. Since the 10th day after the rats were immunized, the weight changes and neurobehavioral scores had been assessed every other morning.After 30 days (remission), the levels of IL-23 and sICAM-1 in serum were measured by ELISA.Results:(1) Through injecting the antigens, the model of EAE was successfully made (2) Neurological function score:In the recurrence period rats (after 18days be modeld), neurological function score of hormone group and electro-acupuncture group was significantly different from model group (P< 0.01). In the remission period rats (after 30days be modeled), neurological function score of hormone group and electro-acupuncture group was different from model group(P<0.05). Electro-acupuncture group compared with hormone group was no significant differences (P>0.05). (3) The level of IL-23:The level of IL-23 was significant different between model group and normal group (P<0.01).The level of IL-23 in electro-acupuncture group and hormone group was similar with that of normal group(P>0.05).But there was different between model group with electro-acupuncture group (P<0.05) and significant different with hormone group (P<0.01). The level of IL-23 was no different between electro-acupuncture group and hormone group (P>0.05). (4) The level of sICAM-1:The level of sICAM-1 was significant different between normal group and model group (P<0.01). The level of sICAM-1 in normal group was similar with that of electro-acupuncture group and hormone group (P>0.05).Al so there was difference between model group with electro-acupuncture groupe(P<0.05). The level of sICAM-1 was different between normal group and hormone group (P<0.05).But the level of sICAM-1 in electro-acupuncture group compared with that of hormone group was similar (P>0.05).Conelusion:Acupuncture treatment and hormone therapy can alleviate inflammation by inhibitting IL-23, sICAM-1 expression Acupuncture treatment and hormone therapy can make IL-23, sICAM-1 keep in normal level in remission; Acupuncture treatment and hormone therapy have the same effect.
