| Lumbricus (also known as earthworm) is a traditional Chinese medicine which has been used for over one thousand years in Chinese history. It has rich resources, significant pharmacological effects, small side effects, low price, and a high value in the clinical application. There are many experiments and studies have showed that lumbricus played obvious role in the promotion of wound healing. It can promote the proliferation of myofibroblasts in granulation tissue, accelerate granulation tissue formation and promote skin growth. Besides, it also can secrete actin, which is conducive to the wound contraction. Therefore, earthworm has been used as a traditional proved recipe to cure burns, scald, lower extremity ulcers and other types of skin wound.It has long been found that the lumbricus has a strong ability of regeneration after the injury. This phenomenon on the one hand is due to that the lumbricus plays a strong role in activating blood circulation to dissipate blood stasis. On the other hand, appropriate stimulation can promote the body to produce Growth Factors and provides nutrients to promote the repair of damaged tissue. Therefore, most terrestrial earthworms can quickly regenerate the removed body segments and restitution. Studies have shown that to break or acupuncture the body of lumbricus can significantly improve the body's platelet-activating factor (PAF).Study on the repair materials produced by lumbricus in the amputated and stimulated state has not been reported in the domestic papers so far. This paper which based on the hypothesis of "Amputated lumbricus" (The regulation of the body's regeneration mechanism will stimulate the body to secrete substances for the body repair after the lumbricus is amputated, and these substances are higher than their normal levels) mainly studied the differences of endogenous materials between the normal lumbricus and amputated ones and explored the relationship between materials differences and the promotion of wound healing activity with the experiments of vivo pharmacodynamics. This experiment used MTT method to select the components that can promote cell proliferation with the study of vitro NIH3T3 cells. It lays foundation for the further elucidate of the mechanism and also provides a new direction for the clinical treatment of trauma and the development of drugs.1,Extract from the broken body of fresh earthworm at different time points were compared by SDS-PAGE technique. The study shows that there are significant differences exist in both the type and content of protein in the extract between the amputated lumbricus and the unbroken one.
