1. ObjectiveTo study the active components alignment of traditional Chinese medicine Radix Puerariae Lobatae compatibilities according to its different functions and indications under the instruction of the active components alignment (ACA) theory. To explore the effective method of quality standards assessment of TCM. To verify the scientific idea that the ACA is TCM standard substance and to establish the new system of active components alignment quality criteria. To provide scientific data and basic methods for establishing TCM quality evaluation method which contains the characteristics of clinic curative effect properly and accurately2. Contents and Methods2.1 Determination of Radix Puerariae Lobatae and preparation of the test productsPrepared drug in pieces of Radix Puerariae Lobatae (guang) and its compatibilities were selected according to the inherited record of traditional clinical medicine combined with the theory of compatibility. Determined the preparation of the test products based on the dosage of compatibilities in agreement with the preparation of traditional medicine.2.2 Pre-screening of the ACA of Radix Puerariae LobataeThe components and pharmacological actions of Radix Puerariae Lobatae were preliminaryly screened through reported documents in which containing Water-soluble compounds, pharmacological actions of compounds related to functions and indications are clear, definite and outstanding and components had more research and good clinical applications.2.3 Studying on the ACA of Radix Puerariae LobataeDtermination and proportion variation ranges of the ACA in Radix Puerariae Lobatae ompatibilities with different functions and indications were analyzed by RP-HPLC based on the decoctions of classics compounds which have definite clinic therapeutic effect.3. Results The ACA of Radix Puerariae Lobatae containing 3'-hydroxy puerarin, puerarin, daidzin and daidzein was determined based on Radix Puerariae Lobatae compatibilities decided by definite clinic therapeutic effect. The ACA of Radix Puerariae Lobatae compatibilities was analyzed and determined by RP-HPLC.3.1 The research simultaneously determined the ACA,3'-hydroxy puerarin, puerarin, daidzin and daidzein in Radix Puerariae Lobatae compatibilities. Peak shapes and separations were preferable, retention times were moderate. The results indicated that puerarin had the highest quantity, then 3'-hydroxy puerarin and daidzin, daidzein had the lowest quantity.3.2 The variation range of the ACA of Radix Puerariae Lobatae compatibilities with different functions and indications was 2.90:5.62:1.30:1-1.50:4.12:1:1, among that the variation range of the ACA of Jiebiao Tuire was 2.90:5.62:1.30:1-2.23:5.19:1.13:1, the variation range of the ACA of xiaoke was 2.43:5.15:1.15:1-1.50:4.12:1:1, the variation range of the ACA of touzhen was 2.09:5.07:1.10:1, the variation range of the ACA of zhixie was 2.67:5.22:1.09:1.4. InnovationDetermined the ACA of Radix Puerariae Lobatae with its main functions and indications according to the theories of compatibility and ACA of TCM. To provide scientific data for the interpretation of the clinic curative effect nature of traditional Chinese medicines in a scientific way and for the studying of the ACA new medicines.
