Cardiovascular diseases based on Atherosclerosis seriously endanger human health. Therefore, prevention and effective control of the progress of the disease, has been to the field of cardiovascular research focus,it also is the focus of attention the world medical proble.Modern medicine referred to in atherosclerosis is closely related with the bleeding, emphasis on blood circulation in the treatment. Access to a number of important ancient Chinese medicine prescription, herbal combination of ancient and mentor for many years of clinical practice, found that many recipes involve blood circulation is commonly used Chinese medicine turmeric turmeric, Curcuma, turmeric. Further literature review found that they all contain one common for the main component of curcumin, therefore, we further studied the effect of curcumin in anti-atherogenic role.This thesis includes literature review and experimental research two parts. the literature review section summarizes the relevant discourse of blood stasis and modern medicine atherosclerotic disease, consolidates the overview of clinical application in the area of bood circulation of Turmeric,Turmeric, Curcuma written in some ancient Chinese medicine and ancient herbal books, also summarizes the basic properties of curcumin, the main pharmacological effects and the anti-atherosclerosis and other aspects of the research summary and progress at home and abrosd in recent years. Study consists of determination of curcumin doses and anti-atherosclerotic effect and mechanism of curcumin two parts.Consolidate the application of blood circulation of turmeric in the 28 dynasties in ancient Chinese medicine books in literature review part,find that there are 36 involved in turmeric, of which 15 under for the treatment of acute heart paralysis, Xiong Xie pain, abdominal pain and other heart; Curcuma involved are 30, including 10 for the treatment of dedication heartache, pain and other confidants; There are 21 involved in turmeric, of which 16 for the treatment of abdominal pain, chest tightness, pain and other confidants. Results can be seen from the above, disorders of clinical application of blood circulation of Turmeric include cardiovascular disease in modern medicine, blood stasis of women's menstruation, prenatal, postpartum in these periods.We selected 20 ApoE (-/-) mice aged 6 week old and randomly divided them into control, curcumin large, medium and low-dose group 4 and fed them with high fat diets in the comfirmation of the dose of curcumin. Adaptive feeding a week later, curcumin large, medium and small dose group were fed curcumin 240mg/kg/d,160mg/kg/d,80mg/kg/d, for 4 weeks.After autopsy,detect plasma TC, TG, HDL-C level; frozen sections of HE staining to observe the right carotid artery adventitial infiltration of inflammatory cells; Sirius red dye sets of Victoria blue method to observe the right carotid artery collagen content, elastic fiber grade and spacing change of elastic fibers; Immunofluorescence to determine the right common carotid artery superoxide dismutase SOD2 expression.The results show that (1) The plasma total cholesterol and triglyceride levels were not significantly different (P>0.05), but compared with other groups levels of HDL was significantly increased in large dose (P<0.05); (2) Adventitial inflammatory cells in each group were gradeⅡinfiltration, no significant difference between types of inflammatory cells (P>0.05), but compared with other groups the number of inflammatory cells of large dose were significantly lower; (3)Ⅰ,Ⅲcollagen showed no significant difference in each group type (P>0.05); Elastic fiber lesions in each group are mainly of gradeⅠ, and the lesion was no significant difference among groups (P>0.05), compared with other groups large doses was significantly reduced elastic fiber spacing (P<0.05); (4) the fluorescence intensity of SOD2 superoxide dismutase of large dose were significantly higher than the other groups (P<0.05).Therefore, we determined to choose the official use of large dose of curcumin.We selected 40 ApoE (-/-) mice aged 6 week old and randomly divided them into normal group, model group, atorvastatin group, curcumin group. Normal group was given to normal diets, the other 3 groups were given to high-fat diets, adaptive feeding a week later, Curcumin group were fed curcumin 240mg/kg/d, atorvastatin group were fed atorvastatin 3mg/kg/d, the normal group and model group were fed normal saline 0.2ml/kg/d,for 4 weeks. After autopsy,detect plasma TC, TG, HDL-C level; paraffin section of HE staining to observe the right carotid artery adventitial infiltration of inflammatory cells; Immunohistochemistry to determine the right common carotid artery inflammation regulating factors NF-κB and inflammatory factor ICAM-1, CD68 expression; Western blot method to determine anti-inflammatory factor aortic PPAR-γ, SOD2 expression. The results show that (1) The plasma total cholesterol and triglyceride levels were not significantly different (P>0.05), but compared with other groups levels of HDL was significantly increased in curcumin group (P<0.05); (2) Adventitial inflammatory cells in each group were gradeⅡinfiltration, no significant difference between types of inflammatory cells (P>0.05), but compared with other groups the number of inflammatory cells of curcumin group were significantly lower (P<0.05); (3) compared with the control group and model group, NF-κB, ICAM-1, CD68 expression showed curcumin group and the atorvastatin group were significantly lower (P<0.05), compared with atorvastatin group curcumin group was significantly lower (P<0.05); (4) compared with model group, other groups PPAR-y, SOD2 protein expression were significantly increased (P<0.05).The results show that curcumin has anti-early-atherosclerosis role in ApoE (-/-) mice,and may be regulated by increasing PPAR-γand SOD2,reducing oxidative stress, thus interfering pathway of NF-κB.
