This study,we discuss the effects of prescription for Yangxue Qufeng on tic behavior and dopamine D2 receptor protein expression in brain of the rat model of chronic tic disorder. And we examine the treatment of children with tic disorders, try to understand the possible mechanism of this prescription. From this research we want to provide some support for the clinical application and experimental basis. This paper includes two parts:review and experimental study.Review includes:"children with tic disorders overview of modern medical research" and"children with tic disorders in Chinese medicine research progress". By searching the relevant literature of tic disorder at home and abroad in recent years,we made a systematic conclusion of modern medicine in children with tic disorder epidemiology, pathogenesis, diagnostic criteria, treatment, animal models, and traditional Chinese medicine in the etiology and pathogenesis, treatment and prescription, experimental study of Chinese medicine.Study:Objective:By establishing a mouse model of chronic tic disorder, observed the behavioral changes and test the content of dopamine D2 receptor protein in brain,we evaluated the animal models, discussed the possible mechanism of prescription for Yangxue Qufeng in children with tic disorders, and compared the pharmacodynamics with tiapride.Methods:140 mice were randomly divided into normal group and model group. The mice of model group gived IDPN by intraperitoneal injection and filtered 70 mice of successful model and 14 normal mice.Divide these mice into normal grup, model group, high dose group of Chinese medicine, dose group of Chinese medicine, low dose group of Chinese medicine and tiapride group. After 4 weeks, compared the behavioral changes and the content of dopamine D2 receptor protein in brain of different groups. Results: The spontaneous activity and nodding movements in the mice which were induced by IDPN increased significantly than those in normal mice. After 4 weeks treatment, the nodding movements in therapy groups were improved to varying degrees. But the result of spontaneous activity in those 6 grups showed no significant difference. The climbing behavior of those 6 grups showed no significant difference before and after treatment. After 4 weeks of medicine therapy, the high dose group of Chinese medicine, dose group of Chinese medicine, low dose group of Chinese medicine and tiapride group compared with the model group, dopamine D2 receptor protein content in brain were significantly reduced. After 4 weeks, the high dose group of Chinese medicine, dose group of Chinese medicine compared with the model group, the food intake of mice were significant increase. But the food intake between Chinese medicine grup and tiapride group showed no significant difference. Conclusion:IDPN-induced mice manifested an increase in nodding movement. The prescription for Yangxue Qufeng can control the mouse's nodding movement and can decrease the dopamine D2 receptor protein in mice's brain. Infer the the possible mechanism of this prescription is the role of dopamine D2 receptor expression down. Thus,it can affects the dopamine system and control the tic symptoms of model mice. The prescription for Yangxue Qufeng can improve the food intake of the model mice of chronic tic disorder.
