Objective:To investigate the effects of celecoxib and its synergistic effects with IFN-αon k562 cell proliferation ,cell cycles,apoptosis and the expression of CD117.Methods:k562 cells were treated with IFN-α,celecoxib and IFN-αcombined celecoxib respectively at different concentrations.MTT assay was used to test the effect of cell proliferation and cell apoptosis , cycles and CD117 expression were determined by flow cytometry .Results:Celecoxib inhibited proliferation of k562 cells ,which was concentration dependence .72h after culture, the rate of k562 cell proliferation in control group, IFN-αgroup , celecoxib group and IFN-αcombined celecoxib group were 96.1%±0.5%,90.2%±0.4%,57.2%±0.9% and 21.9%±0.3% ,respectively. The cell apoptosis rate in four groups were 5.5%±0.8%,6.3%±0.6%,26.4%±3.9% and 57.3%±4.5%,respectively. The CD117 expression rate in four groups were 54.7%,10.5%,36.3% and 7.3%, respectively. IFN-αcombined celecoxib may block k562 cell in G0/G1 phase . Conclusion: Both celecoxib and IFN-αmay inhibit k562 cell proliferation, induce apoptosis , reduce CD117 expression and produce G0/G1 phase blocking in different level and the two drugs has a synergistic effect on the effects mentioned above. Objective:To investigate the effect of celecoxib combining with IFN-αon the BMI-1, CD7 and CD117 gene expression of k562 cells .Methods:k562 cells were treated with IFN-α,celecoxib and IFN-αcombining with celecoxib at different concentrations. Leukemia-related genes-- BMI-1, CD7 and CD117 mRNA expression were detected by real-time quantitative RT-PCR .Results:IFN-αcombining with celecoxib decreased the expression of BMI-1, CD7, CD117 genes mRNA significantly when comparing with the control group (P<0.05).Conclusion:Celecoxib combining with IFN-αhas a synergistic effect on killing k562 cells and the two drugs have a synergistic action on inhibiting the expression of BMI-1, CD7 and CD117 genes.
