| Objective:To investigate the effect of arecoline on insulin resistance induced by the high fructose and fat and explore the potential mechanisms.Methods:Fifty male Sprague-Dawley rats were randomly divided into two groups:the normal control group (n=8), the high fructose and fat group (n=42). Insulin resistance rats induced by the high fructose and fat diet for 12 weeks were randomly divided into 4 groups including arecoline 0 mg/kg,1 mg/kg,5 mg/kg and 10 mg/kg groups (n=8). After 4 weeks, the rats were died. Insulin sensitive index (ISI) was calculated respectively. The content of visceral adipose depots and the shape of fat cells originating from visceral adipose were observed respectively. The mRNA expression of tumor necrosis factor-a (TNFa), IL-6, MCP-1, adiponectin and HO-1 in the visceral adipose tissue were detected by RT-PCR. The protein expression of GLUT4 in skeletal muscle and HO-1 in the visceral adipose tissue were analyzed by western blotting.Results:Arecoline increased singificantly ISI, reduced remarkably the content of visceral adipose depots and the size of fat cells originating from the visceral adipose tissues in the SD rats which were treated with Arecoline (5 and 10 mg/kg) compared with the rats fed with the high fructose and fat diet. The mRNA expressions of TNFa, IL-6, MCP-1 in visceral adipose were decreased, especially in Arecoline 5 mg/kg group. At the same time, the mRNA expression of APN and HO-1 in the visceral adipose tissue were up regulated by 5 and 10 mg/kg Arecoline compared with the rats fed with the high fructose and fat diet. The protein expression of GLUT4 in skeletal muscle and the protein expression of HO-1 in the visceral adipose tissue were increased significantly in arecoline 5,10 mg/kg group compared with the rats fed with the high fructose and fat diet.Conclusion: Arecoline improves insulin resistance induced by the high fructose and fat in SD rats, the mechanism of which may be associated with regulating the expressions of inflammation factors, adiponectin and HO-1 in the adipose tissue and glucose transport 4 in the skeletal muscle by Arecoline... |
