OBJECTIVE:①To observe the protective effects of hongshen chongcao gelatin capsule on diabetic kidney in rats and its mechanism.②To approach the kidney ultramicrostructure change of animal model.METHODS:Experimentation was completed in Yanbian University pharmacy animal laboratory from 2008-6 to 2009-12.①Total 55 Wistar male rats that they were clearing grade, healthy and 8 weeks age, were used in the experiment. The rats were randomly divided into 4 groups:normal control group (n=10),model group (n=15), benazepril-treated group (n=15) and traditional Chinese medicine-treated group (n=15).Rats of normal control group were intraperitoneally injected of natrium citricum buffer solution of same volume;Rats of others groups were intraperitoneally injected of 50 mg/kg streptozotocin which was dissolved by 0.1mol/L citric acid buffer solution before use(pH4.2).Two make model and after 72 hours passed,veinous blood was collected from the tails of models to measure the levels of fasting blood glucose and urine glucose.Blood glucose≥16.7mmol/L and urine glucose over +++suggested that the diabetic models were successful. Rats of benazepril-treated group were performed by intragastric administration of benazepril suspension of 15 mg/(kg·d);Rats of traditional Chinese medicine-treated group were performed by intragastric administration of hongshen chongcao gelatin capsule suspension of 2 g/(kg·d);Rats of nomal control group and model group were performed by intragastric administration of the same volume of running water on each morning, once a day,12 weeks in total.②Levels of blood glucose, blood fat and kidney weight were measured after 12 weeks;③Levels of the kidney VEGF,ColⅣprotein expression was measured by immunohistochemical method;④Use PAS dyeing to observe and compare rats'kidney tissue change;⑤The statistical treatment:To deal with all data by SPSS 16.0 software. The measurement data expressed by mean土S.D. The interclass discrepancy compared by two samples mean comparative T-test, above two samples adopted single factor analysis of variance. The significance expressed by P value:P<0.05 was significance; P<0.01 was very significance.RESULTS:7,5, and 2 rats died in the process of experiment respectively, in normal model group, model group, benazepril-treated group and traditional Chinese medicine-treated group, finally 10,8,10 and 13 rats entered the stage of result analysis.①Weight was significantly lower in the nomal control group(P<0.05,P<0.01):Fasting weight in benazepril-treated group and the traditional Chinese medicine-treated group was significantly higher than model group(P<0.05,P<0.01):There is no weight change in benazepril-treated group and traditional change medicine-treated group(P>0.05);②Fasting blood glucose was significantly higher in the model group than in the model control group(P<0.01),but was significantly lower in the traditional Chinese medicine-treated group than in the model group and benazepril-treated group(P<0.01);③The pathological and formal change of kidney tissue is appeared obviously in the model group, and little change in the beazepril treated group and traditional change medicine-treated group;④Col IVprotein expression in the model group was significantly higher than the normal control group(P<0.01), benazepril-treated group and traditional change medicine-treated group was significantly lower than the normal control group(P<0.01), but higher than normal control group(P<0.01).⑤VEGF protein expression in the model group was significantly higher than the normal group(P<0.01), benazepril-treated group and traditional change medicine-treated group was significantly lower than the model group(P<0.01), but higher than normal control group(P<0.01)CONCLUSION:Hongshen chongcao gelatin capsule restrains on the kidney VEGFand ColⅣprotein expression of rats which were suffered from diabetic nephopathy, therefore decreases desposition of the cell's exterior stroma and works on the protection of kidney. |
