| Objective:To investigate the roles of the MAPK-ERK siganal in proliferation and MMP-2,MMP-9 expresson of gastric carcinoma cell SGC-7901,the specific MEK inhibitor PD98059 was taken to the cells ,inhibiting the activation of the MAPK-ERK signal.Methods:The SGC-7901cells were incubated with different concentrations(0,5,10,20,40um separately)of PD98059 for different times (12,24,48,72hours separatedly);MTT was taken to test the proliferatin; Immunocytochemistory was taken to test the expression of p-ERK;Semi-quantity RT-PCR was taken to test the expressions of mRNA of MMP-2and MMP-9.The cell apoptosis was measured by flow cymetory using anexin V/PI.Results:when SGC-7901 cells were treated with PD98059:1. The cell proliferation was inhibited (P<0.05),and within the experiment span,the effect of proliferation inhibition was increasd with the concentrations(P<0.05)of PD98059 and interfering times(P<0.05);2.p-ERK expression was suppressed; 3.The cell apoptosis percentage was increased obviously.4.The expression of mRNA of MMP-9 was decreased(p=0.000), and meanwhile the expression of mRNA of MMP-2 was not obviously interfered (p=0.118);Conclusions:1.The activation of MAPK-ERK signal is inhibited after the SGC-7901cells are treated with PD98059;2.PD98059 can inhibit the proliferation and promote apoptosis.3.MAPK-ERK signal takes part in modulating the MMP-9 expression of SGC-7901cells,and the expression can be inhibited by suppression the activation of the signal. |
