Objective: By observing the expression of retina' neuroglobin and cornea' transforming growth factor-82 at plateau and high- altitude after ocular contusion, to investigate the relationship between the ocular reaction after contusion with hypoxidosis at high- altitude and the two factors, in order to offer a therapy threshold of clinical anoxic ocular disease.Methods: 50 (n=100) rabbits were randomized into 4 groups, un-injurygroup (eyeballs without injury, in the plateau and under the high-altitude), injury group (eyeballs with injury, in the plateau and under the high-altitude), ocular contusion models were established in the last two groups(both eyeballs were to be injured), in which underwent damage for 2nd hour, 6th hour, 1st day, 3rd days, 7th days, 10th days, 14th days. All the eyeballs were taken out at different time. NGB in the retina were detected by using histology and immuno-histochemical techniques respectively.Results: The development variety of NGB-IR(NGB-immunoreactive) is similar between plateau group and high- altitude group, it increases with time in the 4 times from the 2nd hour to the 3th day, and reaches to the peak on the3th days.After 7th day,the expression of NGB begin to decline. Both high-altitude group and plateau group is higher than the un-injury group of NGB-IR between the 7 times (P<0.05) .Although high-altitude group is high compared to the plateau group , especially at the2th hour (P<0.01),the increasing rate of NGB in plateau group is higher than that in high-altitude group.The difference of the expression of TGF-β2 between high-altitude group and plateau group is significant, especially in 6h, 1d, 3d, for in all these times, the expression of TGF-β2 the plateau group is higher than in high-altitude group.In Conclusion: (1) The expression of NGB in the retina under high-altitude is higher than that in the plateau . (2) After ocular contusion, the expression of NGB in the retina under high-altitude is higher than that in the plateau. (3)After ocular contusion, the increasing rate the expression of NGB in the retina in the plateau is higher than that under the high-altitude. (4) After ocular contusion, the expression of TGF-β2 in the cornea in the plateau is higher than that under high-altitude.Conclusion: (1)The expression of NGB in the retina under high-altitude is higher than that in the plateau . (2)After ocular contusion, the expression of NGB in the retina under high-altitude is higher than that in the plateau. (3)After ocular contusion, the increasing rate the expression of NGB in the retina in the plateau is higher than that under the high-altitude. (4)After ocular contusion, the expression of TGF-β2 in the cornea in the plateau is higher than that under high-altitude. (5) After ocular contusion, the structure of mitochondrion in the retina is damaged. |
