Objective: To investigate the change of iron metabolism in rats, We established the rat model of PTE by injecting FeCl3 to the cortex, then used DFO to evaluate whether there was an inhibitory effect of DFO on PTE.Methods:1. Forty male Sprague Dawley rats were randomly divided into the normal control group, the surgery control group and PTE model 24 h, 5 d, 15 d groups with 8 rats respectively. We established the rats PTE model by the micro-injection method. By using the methods of flame atomic absorption spectrometry (FAAS) and enzyme-linked immunosorbentassay(ELISA), we detected the contents of iron and Tf in the brain of rat models and explored the role of iron in PTE.2. Seventy-two male Sprague Dawley rats were randomly divided into the PTE model 24 h, 5 d, 15 d groups, the DFO pretreatment 24 h, 5 d, 15 d groups and the DFO early intervention 24 h, 5 d, 15 d groups. We established the rats PTE model by the micro- injection method and gave the DFO immediately or 12 hour's later respectively to observe whether DFO had the suppressive effects.Results:1. The change of iron metabolism in PTE model brain: In the entire experiment cycle, the iron content which the injection side of brain of the model demonstrated the declining trend, but was higher than the normal control group.2. The frequency of seizures after DFO treatment: In the entire experiment cycle, DFO pretreatment significantly decreased the frequency of seizures by 71.11% in the DFO pretreatment rats compared with that of the PTE model group. There was significant difference between the two groups(P<0.05). And DFO later intervention also decreased the frequency of seizures by 30.31% in the DFO early intervention rats compared with that of the PTE model group. There was significant difference between the two groups(P<0.05). Between DFO pretreatment and DFO later intervention, the frequency of seizures was significant difference.3. The change of iron content after DFO treatment: 24 hour's later after the PTE model successfully establishment, we found that there was significant difference of the rat brain iron content in PTE model group both with the DFO intervention group and the DFO early intervention group(P<0.05). But there was no statistical difference of the rat brain iron content between the DFO pretreatment group and the DFO early intervention group. 5 d and 15 day's later after the PTE model successfully establishment, we found that there was no statistical difference of the rat brain iron content in the PTE model group both compares with the DFO pretreatment group or with the DFO early intervention group.4. The change of Tf content after DFO treatment including: When we injected DFO immediately after PTE model successfully, we found that there was significant difference of the rat brain Tf content in DFO pretreatment 24 h group both compares with the DFO pretreatment 5 d group and with the DFO pretreatment 15 d group (P<0.05). However, there was no statistical difference of the rat brain Tf content between DFO pretreatment 5 d group and DFO intervention 15 d group. Each point of the study, there was significantly difference of the rat brain Tf content in DFO early intervention groups(P<0.05).Conclusion:1. The content of iron and Tf are increased in rats brain, which indicates Iron may be a part of the mechanism of PTE, moreover is possibly the onset factor of PTE.2. Treatment with DFO in the PTE model rats, either pretreatment or early intervention may reduce the times of seizure, especially the pretreatment is more effective. It also reduces the content of free iron directly which suggest that DFO may be used for PTE therapy.
