| Objective:To observe the effect of Naoling Decoction (NLD) on ethology, hippocamal histomorphology, and expression of BDNF and TrkβmRNA in hippocamp of the rats with synthetic Alzheimer's disease, and explore the therapeutical effect on Alzheimer's disease in rats and the potential mechanism of Naoling Decoction.Methods:30 pure line SD (spragu-dawleg) rats were classified into five groups:normal group, sham- operated group, model group, Naoling Decoction group and Piracetam group. The model with Alzheimer's disease was established by Aβ1—42 injected into hippocamp in rats. The faculty of learning and memory of the model rats was evaluated by Y-electric-maze test and Morris water maze experiment after 4 weeks intragastric administration. The brain tissues were sliced into frozen sections. The expression of BDNF and TrkβmRNA in hippocamp was measured with hybridization in situ.Results:(1)The results of Y-electric-maze test showed that compared with sham-operated group and normal group, the frequency of evading stimulation of model group is increased obviously (p<0.05); compared with model group, the frequency of evading stimulation of Naoling Decoctio group and Piracetam group is decreased (p<0.05). (2) The results of Morris water maze experiment showed that compared with sham-operated group and normal group, the escape latent period of model group was delayed significantly (p<0.05). In exploratory experiment, the average pass through frequency in sham-operation group and normal group was most, and that of the model group was less than that of DLC group and Piracetam group significantly (p<0.05). (3)The results of hybridization in situ of BDNF showed:The compasion of the gray scale value about BDNF in hippocamp of each group, the model group were significantly higher than sham-operated group and normal group (p<0.05); the gray scale value of Naoling Decoction group and Piracetam group were lower than the model group's gray scale value significantly (p<0.05). (4) The results of Hybridization in situ of Trkp mRNA showed:The compasion of the gray scale value about TrkβmRNA in hippocamp of each group, the model group were significantly higher than sham-operated group and normal group (p<0.05); the gray scale value of Naoling Decoction group and Piracetam group were lower than the model group's gray scale value significantly (p<0.05).Conclutions:(1)The model which was established by Aβ31-42 injected into hippocamal was the best, because the model rats had some praxiological and pathological changes. (2)The learning and memory of the model rats were decreased obviously, which indicates that Naoling Decoction can improve the ability of learning and memory of the model rats. (3)Naoling Decoction can enhance the expression of BDNF and TrkβmRNA in hippocamp of AD rats, which may be the some part of therapeutic mechanism on Alzheimer's disease of Naoling Decoction. |
