Objective:To observe the changes of eosinophils in peripheral blood and tissues of stomach and intestine, validating that EG model was made for success and it can be as the Prescription-syndrome model of banxiaxiexin decoction; to observe the effects of banxiaxiexin decoction act on the expression level of Eotaxin-1,studying it can cure the Eosinophilic Gastroenteritis by the path of CC chemokine receptor 3.Methods:We prepare Eosinophilic Gastroenteritis mice models. Treatment groups must be gavaged by banxiaxiexin decoction,prednisolone and xiangshaliujunzi decoction after making models,all the mice were sacrificed after twenty days.we take acount of Eosinophil through the peripheral blood.we make the pathological section,noemal HE staining and observing the pathological morphologic change and taking acount of Eosinophil.we detect the contents of immunoglobulin A and immunoglobulin G by the Enzyme-Linked Immuno Sorbent Assay after collecting the flushing fluid of small intestine.Other fresh gastric tissue should be frozen reserved at -80℃.We adopt the method of FQ-PCR detecting the expression level of Eotaxin-1 (EOT-1). The datum was analyzed by SPSS 16.0.Results:1.The count of Eosinophils:The count of eosinophils in model group was higher than normal group respectively, P<0.01.Among the therapy groups,the count of eosinophils in banxiaxiexin decoction group and prednisolone group was lower than model group respectively,P<0.05,but these were no significantly difference among the model group and xiangshaliujunzi decoction, P>0.05.2.Patholog:Mucosa covering the Eosinophilic Gastroenteritis mice model control group is little,which is granulation tissue of shallow layer,The gap is covered by little generated mucosa,and its gland is arranged in disorder,appering cystic dilatation,especially the increased count of Eosinophil i(?) the small intestine tissue.Among the therapy groups,mucosa of ulcer surfaces in banxiaxiexin decoction and prednisolone group have formed epithelium,lamina propria muscularis and mucosa plica.Mucosa covering on ulcer surface in xiangshaliujunzi decoction group have formed complete epithelium and lamina propria muscularis,though the depth of mucosa is a little thiner, the forming morphology of plica is better than model control group significantly.3.Detecting the contents of immunoglobulin A and immunoglobulin G in the flushing fluid of small intestine:There were significantly difference among the contents of immunoglobulin A and immunoglobulin G in model group and normal group,P<0.05. Among the therapy groups, Among the therapy groups,the differences between banxiaxiexin decoction group and prednisolone group have no statistical significance,P>0.05.Compared banxiaxiexin decoction group and prednisolone group to model group, they have significant difference,P<0.05. The differences between banxiaxiexin decoction group and prednisolone group and xiangshaliujunzi decoction group have no statistical significance,P>0.05.but the differences have statistical significance in the comparing of xiangshaliujunzi decoction group and model group,P<0.05.4.The expression level of Eotaxin-1:The expression level of Eotaxin-1 in banxiaxiexin decoction group and prednisolone group was lower than model group respectively,P<0.05. The differences of the expression level of Eotaxin-1 between model group and xiangshaliujunzi decoction group have no statistical significance, P>0.05.Conclusion:1.The EG model which was based on being gavaged by PLGA+OVA and OVA+AL(OH)3CI. is successful.2.The differences of clinic symtom, Patholog change, decrease of EOS and IgA, IgG and the expression level of Eotaxin-1 between banxiaxiexin decoction group and prednisolone group have no statistical significance,P>0.05.So, the EG model can be as the studying model of banxiaxiexin decoction.3.The level of immunoglobulin A and immunoglobulin G in the flushing fluid of small intestine was decreased by banxiaxiexin decoction, this may be proved that banxiaxiexin decoction could repair the mucous membrane by that ways.4.The expression level of Eotaxin-1 and the number of EOS in banxiaxiexin decoction group were depressed, however the action of Eotaxin-1 can be only on CCR3, so we can say that the path of therapy in EG may be pass the molecular mechanism of CCR3.
