Objective:1.To investigate the distribution of IL-6 gene-174G/C and C-reactive protein(CRP) gene +1059G/C polymorphisms between MS group and control group in Xinjiang Kazakh and Han race. 2.To study the associations of the gene polymorphism at-174G/C interleukin-6 and C-reactive protein (CRP) +1059 G/C in patients with metabolic syndrome and its components of Xinjiang Kazakh and Han race.Method:1. Stratified cluster sampling method was used to cross-sectional investigation among the local Bole and Shawan residents aged≥18 from April to May of 2008,200 patient with MS were selected,which suggested by International Diabetes Federation (IDF) and at the same time 201 without MS controls were randomly selected of Kazakh and Han population.2.A polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) was used to detect the frequency of IL-6-174G/C and CRP+1059G/C genotype and alleles of Kazakh and Han residents.3.Theχ2 test was adopoted to check the difference in genotype distribution between MS cases and controls, and the accordance with Hardy-Weinberg equilibrium in the group. All the calculations and statistics were performed with the computer program, SPSS Version 13.0. Statistical analysises adopt t test,χ2 test, odds ratios and 95% confidence intervals for possible risk/protective factors.Result:l.The genotype distributions of IL-6-174G/C and CRP+1059G/C were in accordance with Hardy Weinberg equilibrium.2.The genotype and allele frequency distributions of IL-6-174G/C:(1) Through PCR-RFLP genotype detection,there are three genotypes in Kazakh race:GG, GC and CC, the frequency of GG genotype are far high than GC and CC genotypes; the frequency of GG genotypes is the highest, and that of G alleles is obviously higher than that of C allele in Kazakh and Han race,but CC genotype was found in Kazakh race; Frequency of GG genotypes were 94.00% and 98.50%,GC were 6.00% and 1.50% in Kazakh two groups, G and C allele frequency were 97.00% and 3.00% in MS group; G and C allele frequency were 99.30% and 0.70% in control group, there was significant difference between MS group and control groups (χ2=5.66, P<0.05); The frequency of GG genotypes were 93.43% and 97.51% GC genotypes were 6.57% and 2.49% in Han MS groups and control groups; G alleles frequency were 96.71% and 98.76%, C alleles frequency were3.29% and 1.24%, there was significant difference between MS group and control groups (χ2=4.52,P<0.05); but the genotype and allele frequency distributions of IL-6-174G/C and CRP+1059G/C were no significant difference in Kazakh and Han race(P>0.05).3. The genotype and allele frequency distributions of CRP+1059G/C:(1) Frequency of GG genotypes is the highest, and that of G alleles is obviously higher than that of C allele in Kazakh race, CC genotype was no found in Kazakh race; Frequency of GG genotypes were 93.0% and 99.0%, GC genotypes were 7.0% and 1.0% in MS groups and control group. The frequencies of genotype and allele between MS group and control groups were significantly different in Kazakh race(χ2=9.43, P<0.05);4. Association of IL-6-174G/C and CRP+1059G/C with metabolic syndrome and its components in Kazakh and Han race.:(1) In IL-6-174G/C:GG genotypes its WC,TG,HDL-C,SBP,DBP comparison with and control groups difference has statistics significance (P<0.01) between MS group and control group in Kazakh race, GC genotypes its WC,HDL-C comparison with and control groups difference has statistics significance (P<0.00);GG genotypes its WC,HDL-C,SBP,DBP,FPG comparison with and control groups difference has statistics significance (P<0.05), between MS group and control group in Han race, GC genotypes its WC,TG,SBP comparison with and control groups difference has statistics significance (P≤0.05); (2) In CRP+1059G/C:GG genotypes its WC,TG,HDL-C,SBP,DBP,FPG comparison with and control groups difference has statistics significance (P<0.01),GG genotypes its WCcomparison with and control groups difference has statistics significance (P<0.01) between MS group and control group in Kazakh race.5. Association of GG and GC genotypes at IL-6-174G/C with metabolic syndrome abnonnal components gather in Kazakh and Han race:GG and GC genotypes with metabolic syndrome abnormal components gather number are no evident change,GC genotypes was opposite in Kazakh race, there was significant difference between GG and GC genotypes with metabolic syndrome abnormal components gather number (χ2=18.45, P=0.00); Similar change were also reflected in Han race. there was significant difference between GG and GC genotypes with metabolic syndrome abnonnal components gather number (χ2=16.34, P=0.006)Conclusion:1. These results suggest that the interleukin-6 gene-174G/C and CRP+1059 polymorphism was associated with metabolic syndrome and its components of Xinjiang Han and Kazakh race.
