Objective:To investigate the effects of Haishengsu on the proliferation and the mechanism on K562 and K562/ADM cells, to study the effects of inducing apoptosis and restraining cell cycle, to detect the expression of gene and protein with Sorcin and mdrl in K562/ADM cells.Methods:The procedure of MTT assay was used to determine the restraint in K562 and K562/ADM cells, immunocytochemistry was used to test the expression of bcl-2 and bax protein, FCM was used to investigate the early apoptosis ratio and cell cycle and the expression of Pgp,SDS-PAGE and Western blot were used to explore the expression of Sorcin.Results:Haishengsu inhibited the proliferation of K562 cells significantly on the concentration 20μg.ml-1,it could inhibite the proliferation of K562/ADM cells with 40μg.ml-1; the ratio of inhibition on the proliferation was higher statistically than control (p<0.05),and it related to the time and concentration; Haishengsu blocked the phase G1/S of cell cycle in K562 cells; Haishengsu induced apoptosis in K562 and K562/ADM cells, the apoptosis ratio was (10.53±2.45)% for 12 hours on the concentration 20μg.ml-1 in K562 cells, it was higher than control significantly, with the extension of time and increasment of concentration, the most amount of K562 cells was dead; An extra Sub-G1 peak ahead of G1 peak on DNA histogram was detected by FCM after imposed on K562/ADM cells with Haishengsu on the concentration 40μg.ml-1. The apoptosis ratio was higher than control, it increased with the time. Haishengsu could down-regulate the expression of bcl-2 protein and upregulate the expression of bax protein in K562 and K562/ADM cells, while it could not affect the expression of Sorcin and mdrl genes, but could down-regulate the expression of Pgp in K562/ADM cells.Conclusion:1.Haishengsu could significally inhibite the proliferation of the K562 and K562/ADM cells;2.Haishengsu blocked the phase G1/S of cell cycle in K562 cells;3.Haishengsu could down-regulate the expression of bcl-2 protein and upregulate the expression of bax protein in K562 and K562/ADM cells,and induced the apoptosis of K562 and K562/ADM cells;4.Haishengsu might have influence on drug-resistance of K562/ADM cells through regulating the expression of bcl-2 and bax protein in K562/ADM cells;5.Haishengsu could down-regulate the expression of Pgp and had no effects on the expression of mdrl gene in K562/ADM cells;6.Haishengsu could not affect the expression of Sorcin and mdrl genes...
