Objective: To explore the anti-tumor mechanism of oleanolic acid, the active ingredients of Chinese Ligustrum lucidum on mice in vivo on molecular level.Methods: Establish S180 tumor-bearing mice model to compute the inhibition rate and life prolonging rate ;To observe by protein electrophoresis of tumor protein and serum protein band changes; by immunohistochemistry to observe the cell cycle control genes P53 and tumor necrosis factor P53expression.Results: The oleanolic acid has inhibitory tumor effect. The tumor inhibition rate was 48.59%, the life extension rate was 44.62%, and no obvious side effects.Oleanolic acid some mice tumor protein and serum protein expression affected.Oleanolic acid can be reduced tumor cell expression of mutant P53 protein, increases the expression of tumor necrosis factor, compared witn the model group, the difference was statistically significant (P <0. 01).Conclusion: The active ingredient Ligustrum oleanolic acid can effectively inhibit tumor growth and prolong survival time of tumor-bearing mice. The oleanolic acid can influence the expression of tumor-bearing mice serum protein and tumor tissue protein obviously..Oleanolic acid can affect the cell cycle regulatory genes p53 in tumor cells and tumor necrosis factor expression.The experiment proved that ligustrum lucidum proved effective component oleanolic acid has good anti-tumor effect in vivo.
