Because of increased caloric intake and reduced physical activity,the prevalence of obesity, especially among adolescents,has increased considerably over the past 20years. It has been estimated the 20% of the world's population is overweight and nearly 300 million world population are obese (BMI≥30 kg/m2).In China ,the stress of obesity and overweight is also severely. 22.8% of China's population is overweight and nearly 60 million are obese. Also epidemiology data show that yellow race are sensitively to obesity .Excess body weight and obesity are onset associated with the development of metabolic syndrome and type II diabetes, both of which are closely associated with insulin resistance and increased risk for cardiovascular complications. So,approach the mechanism of obedity , prevention and treatment the obesity-related complication is important and meaningful. Previous studies demonstrated that obesity is associated with activation of inflammatory pathways and that inflammatory responses are associated with impaired insulin signaling and insulin resistance.In the obesity people ,adipose tissue is characterized by increased infiltration of macrophages, suggesting that they might represent an important source of inflammation.And the production of proinflammatory and antiinflammatory cytokines is dysregulated, which may play an important role in the development of obesity-related sequelae.In our study ,omental adipose tissue biopsies were obtained from patients who were undergoing elective open-abdominal surgery.The adipose tissue were cultured and stimulate by interleukin-6(IL-6),the detection of cell proliferation were used the method CCK-8. Afer the proadipocyte were induced differentiation by insulin,dexamethasone,the culture solution was added IL-6,then the level of differentiation were detested with the method of oil red O.The primary culture and differentiated induction of human preadipocytes were performed, and were treated with IL-6 at different concentrations. The expression gene level of the IL-6 receptor—gp130, adiponectin and the gene of peroxisome Proliferator-activated receptor(PPAR-γ) mRNA were also assessed with the method of RT-PCR and real-time PCR.The result show that: IL-6 could markly suppressed the proliferation of preadipocytes. And it could also suppressed the differentiation of adipocyte . After stimulation with IL-6 , gp130 was upregulated and adiponectin and PPAR-γis suppressed.At last we simulated the adipose tissue of obesity by cocultureing the human primary adipocyte and mononuclear macrophage THP-1.The result show that Toll like recepor-4(TLR-4) was increased than that of culture alone.That may indicate that adipocyte and mononuclear macrophagecould interact with each other in the process of obedity and the complications of obesity.
