In this paper,the extracting and purifiacting flavonoids from Cyclocarya paliums ,antioxidative and antibacterial activity were studied. The results were as follows:Design with four factors (ratio of solvent to dry Cyclocarya paliums leaves,extraction time,ultrasonic power and ethanol concentration) as influence factors. The results indicated that the effect of factors on the extraction was in the order as follows, extraction time, the most important one, then ethanol concentration, ratio of solvent to dry Cyclocarya paliums leaves, and ultrasonic power as the last one. The optimum conditions were ratio of solvent to dry Cyclocarya paliums leaves 55:1, extraction time 34min, ultrasonic power 720W, and ethanol concentration 60%. With the optimized technology, the content of total flavonoids in the final product was 86.53%.The optimal technologies of separation and purification of flavonoid were obtained by adsorption efficiency and elution efficiency of static adsorption and elution tests. The results showed that the AB-8 macroporous resin displaved the best effect of separation with the sampling rate and concentration 2BV·h-1 and 1.0 mg·mL-1, respectively, and the concentration of elution was 70% alcohol, with eluting velocity was 2BV·h-1.The results of antioxidative showed that in a certain range, Cyclocarya Paliurus flavonoid extract on superoxide radical (O2-·) role as the removal of the constant increase in the amount of curve and a rise in the1.2 6.0 mg amount, Extracts of the hydroxyl radical (? OH) increased the clearance of-way, but excellent extract is the first increase after declining trend in the removal of H2O2 in the study, refined extract of the clearance rate was significantly higher than that extracts. The study of oil extract of anti-oxidation experimental showed that the antioxidant activity of flavonoids by adding strength and concentration, the higher the concentration, the stronger the antioxidant activity.The results of antibacterial activity showed that the total flavonoids from Cyclocarya paliums leaves had strong anti-bacterial activity against Three types of bacteria , The MIC and MBC of Staphylococcus aureus,Escherichia coli were 40 mg·mL-1, 40 mg·mL-1; 37 mg·mL-1,40 mg·mL-1 of Bacillus subtilis and 25 mg·mL-1,28 mg·mL-1 of Escherichia coli.and the antibacterial effects is better at pH5.
