Belonging to the genus of lactarius, Russulaceae family and Russulales, Lactarius hatsudake Tanaka, a precious indigenous mushroom, is mainly distribute in China, Japan and etc.. Because of its delicious, eutrophy, eatable and pharmaceutical, it is to be in great demand in the areal area. It that lives with especial ecological, nutritional mode and condition to differentiatiate fruiting, which is symbiosis with Pinus massoniana Lamb , so it is unable to cultivated by artificial now. Supply of the market are mainly from wild collection. It leads to serious natural resources destruction because of intemperance in pluck, and Lactarius hatsudake Tanaka output are gradually decreasing. To protect diversity of edible fungus and meet consumers' needs, it is an investigatory focus both here and abroad to adopt submerged cultivation technique for Lactarius hatsudake Tanaka and other domestic funguses .This paper studies wild Lactarius hatsudake Tanaka strain segregation, hypha pure cultivation, hyphal liquid culture conditions, nutrient contents analysis of fruiting and body mycelial, and extraction technics of polysaccharide. Main findings, as follows:1. Chose the PDA medium , and got the strain of Lactarius hatsudake Tanaka successfully used the organism separation and spore separation methods.2. Measured and analyzed the nutrition ingredient of Lactarius hatsudake Tanaka both hymenium and mycelium ,there is no large difference between them.3. Improvement on the condition of the liquid fermentation mycelia and the polysaccharide extractive technique of Lactarius hatsudake Tanaka with the response surface methodology, the result indicate that the most mycelia biomass is 13.17 g/L under the condition of the liquid fermentation mycelia that is : maltose 40.09 g/L\ peptone 3.35 g/L,K2HPO41.98 g/L,temperature 30.04℃;the ratio of material and liquid is 1:35.2, temperature 39.5℃,extracting 34.0min,the most efficiency of polysaccharide can attain 6.61%.4. Reached on the ultrafilitration condition of the polysaccharide extraction of Lactarius hatsudake Tanaka, the most technics condition is :the standard of ltrafilitration film is 5KD,the pressure is 0.1 MPa,the temperature is 30℃.
