| Sedum shitaiense Yan Zheng et D. C. Zhang is one of the common evergreen herbaceous plant that belong to Crassulaceae. It is a superior horticulture plant, which is elegant, well-adapted, beflexible and easy cultivation, ect. And can serve as cover plants or miniascape for decorating flowerbed, courtyard, study room and living room, ect. The peculiar crassulacean acid metabolism of sedum can absorb carbon dioxide and release oxygen in the darkness, increase the concentration of indoor air negative ions. Using as green plant, it has potential for a broad range of application. This paper had studied anatomy and tissue culture of S. shitaiense, which corresponded to the study of horticulture function and industrialization. The results were as following:1. Morphological and Histological Studies of S. shitaienseThe stem structure, leaf epidermis structure and the pollen morphology of S. shitaiense were examined in detail under light microscope and scanning electron microscope. According to the anatomical studies, the stems of S. shitaiense consist of epidermis, cortex and primary vascular tissue. Cortex was relatively advanced, the vascular tissue accounts for the proportion of stems relatively small, and the idioblast is rich in the stem. The shape of surface cells of S. shitaiense was anomaly and the ornamentation was densify. The stomata were anisocytic and distributed on upper and lower epidermis randomly. As far as pollen morphology was concerned, the globelike shape, angulaperturate, 3-colporate and reticulate ornamentation type on the exine surface.2. Tissue Culture of S. shitaienseThe main factors affecting callus induction, bud proliferation and rooting were studied using leaves and stems of S. shitaiense as explants. Result showed that the optimal medium for callus inducement and buds differentiation from leaf segments and stems was MS supplemented with 2.5mg/L 6-BA and 0.3mg/L NAA, and the callus inducement rate reached 90 percent. Yellow and green callus could be induced from leaf segments, but only green callus could be induced to bud easily in the medium. The MS medium supplemented with 2.0mg/L 6-BA and 0.3mg/L NAA was favorable for the proliferation of buds, and the average regeneration plantlets for each explant was 12. 14 after 4 weeks of culture. All the regenerative plantlets could take root after being transferred to 1/2MS medium supplemented with 0.1mg/L NAA.3. The Evaluation of Horticulture Function of S. shitaienseThis part located the horticulture function of S. shitaiense as an ornamental plant which has greening and beautifying effect, and introduced its ornamental charateristics from the plant type, leaf, flower and so on. The result was that the application of analytic hierarchy process on the evaluation of horticulture function of Sedum can well reflect synthesis horticulture function of Sedum.4. The Study on the Industrial Mode of S. shitaienseThe last part researched the industrial mode of S. shitaiense, established the industrial model, and elaborated the ponderation about industrialization development from fast reproductive system of plants' tissue culture and its industrialization and marketing industrialization. The using of tissue culture technology on S. shitaiense can provide a large number of seedlings without territorial restrictions and weatherprotected. |
