| In this experiment, use the Pleurotus eryngii as experimental material, ISSR markers throughout it's traditional genetic cross breeding process to provide the basis for breeding strains of Pleurotus which have good character and study the role of molecular markers in the traditional mushroom cross breeding.The results are as follows:1. the genetic diversity of mushroom species of Pleurotus eryngii from the different regions of China was analyzed with ISSR(Inter-simplesequence repeat) marker. The result showed that 202 bands were amplified with 16 polymorphlc primers including 158 polymorphic ones and their polymorphism percentage was 78%. showing a wealth of ISSR genetic polymorphism; 30 samples could be clustered into 8 groups based on the UPGMA method with NTYS-PC2.1.the full description of this experiment prove that there was more bigger genetic diversity in our country, so as to foundation for screened out the larger GDs species.2. election one from each class as the experimental material, through the growth of mycelium, the agronomic characteristics of fruiting body, yield and polysaccharides and a series of characters compared to filter 3 kinds of high-quality mushroom strains parents, The result showed that X12,X29 and X30 have the best character, were the best choice of the parents.3. the three different varieties of mushroom which have good comprehensive characters is analyzed by mating-type single-spore isolation and ISSR analysis, the results show us that: the results of traditional single-spore isolation method is same with the results of the ISSR analysis, four different mating types occurred in different variation .4. use the different mating types to do the traditional cross-breeding experiments, use the hybrid offspring and their parents do the preliminary comparison of the growth velocity and ISSR analysis. the results show us that: the hybrid offspring of X29 and X30,X12and X30 have better character than their parents,however, the hybrid offspring of X12 and X29 appeared some decline.In summary, this experiment not only obtained 14 hybrids to provide the basis for the identification and selection of superior strain, but also provide the technical support in study mushroom crossbreeding by understanding the growth and development of edible fungi and the ISSR polymorphism analysis. |
