In this paper, the author conducts in-depth research for the optimization of bus stop location about Bus Rapid Transit System. The contents of the paper contain: Analyzing from the new land-use characteristics of China urban development presently, put forward the concept of Trip Generation Source, and draw the conclusion that the relationship between Trip Generation Source and bus stop location of BRT is the Passenger Flow Density. Simultaneity, bring forward the optimization mode of bus stop location based on economic theory and lower money cost for different objects; According to the trip generation forecast methods under the present situation of China traffic data collection, narrate the process of the suitable trip generation forecast method for China-"Trip Generation Rate based on land-use" and set up the improved forecast system based on Trip Generation Source; Analyzing from the detail behaviors about BRT passengers, create the technic of money cost calculation; Under the practice means of SGA, put forward the calculated method- "Random Array Create" to gain the answer of optimization mode and program the computer with language "c" to do the job and then Regard Beijing BRT Line One as the case study to validate these theory.
