This paper selected lead(Pb), cadmium (Cd), m-dinitrobenzene (m-DNB), 2,4-dinitrotoluene (2,4-DNT), 2,6-dinitrotoluene (2,6-DNT), 4-nitrotoluene (4-NT) and 2,4-dinitroaniline (2,4-DNAn) as research objects, using Sertoli-germ cell coculture and single cell gel electrophoresis to study the damage on the DNA of germ cell induced by these seven chemicals at the molecular level of biological toxicity. Moreover, this paper also made a primary discussion on the reproductive toxicity and the mechanism, which provided the basis for the safety estimate of ecological toxicology at the DNA level. The results showed that these seven chemicals had the reproductive toxicity. All of them had lead to the damage on DNA of germ cell of kunming male rat in the definite range of concentration. With the concentration increasing, the damage rate also increased, which showed an evident logarithm dose-effect relationship with regression equations and correlation coefficients as following:Lead: at the concentration of 0.004~lmmol/L, it can lead to the DNA damage of germ cell in rat, regression equation was y=0.26241gx+0.9893, and correlation coefficient is 0.9948.Cadmium: at the concentration of 0.00008-0.05mmol/L, it can lead to the DNA damage of germ cell in rat, regression equation was y=0.1961gx+1.159, and correlation coefficient is 0.9999.m-dinitrobenzene: at the concentration of 0.00004~0.025mmol/L, it can lead to the DNA damage of germ cell in rat, regression equation was y=0.15171gx+1.223, and correlation coefficient is 0.955.2,4-dinitrotoluene: at the concentration of 0.000032-0.5mmol/L, it can lead to the DNA damage of germ cell in rat, regression equation was y=0.07361gx+1.053, and correlation coefficient is 0.9612.2,6-dinitrotoluene: at the concentration of 0.000032-0.5mmol/L, it can lead to the DNA damage of germ cell in rat, regression equation was y=0.09591gx+1.1909, and correlation coefficient is 0.9761.4-nitrotoluene: at the concentration of 0.000032~0.5mmol/L, it can lead to the DNA damage of germ cell in rat, regression equation was y=0.25471gx+1.4407, and correlation coefficient is 0.9760.2,4-dinitroaniline: at the concentration of 0.0008~0.02mmol/L, it can lead to the DNA damage of germ cell in rat, which showed an evident dose-effect relationship.In the regression equations as stated above, x presented dose (mmol/L) and y represented the decimal fraction of damage rate. After the significant test, correlation coeffiency is very significant, p<0.01.
