Trehalose is a kind of indeoxidized disaccharide coupled with tow glucose by a ,a -1,1 bond. Molecular formula is C12H22O11 and molecular weight is 378. 23(with two H2O). In the creature body, trehalose not only is a structure component which provide energy, but also is a metabolized substance which can protect the protein , the fat, the glucide and the nucleic acid in creature cell at dry, low temperature, high pervasive condition against demolishing. Trehalose has great potential for application in food., medical treatment and agriculcure.The nature determination of trehalose through TLC is studied. Finally the experiment separate glucose, sucrose .maltose trehalose by using GF254 TLC board(French). The sample is spreaded in liquid which n-B utyl alcohol:Pyridine:Water is 15:3:3, after second spreading, developing at 105℃ in lipuid H,SO,:OU)=1:4. The sensitivity is 0. 04ug.The experiment also study fix quantify method in lab. The result show that anthrone colorimetric method, optical rotation method and TLC method all can fix trehalose quantify.Microzyme B-15 is filtered from 12 microzyme conserved in laboratory and isolated from hot spring in changbai mountain and fermentive honey. Microzyme ZY-D10, abtained from mutagenizing B-15,contain 155. 23mg/g, which is 0.8 more than B-15.The culture condition of microzyme ZY-D-10 is studied, temperature 32℃, time 27h, PH 6. 5. After culturing, trehalose content in the yeast extract is 170. 7mg/g, which is more 99.4% than the initial strain. The experiment find anther way to produce trehalose through testing yeastiness which has high trehalose content.At the stusy of method of determination the nature and fix quantify for the trehalose this experiment screen the strain which has high production from 16 kinds of yeasts. The research filtered the strain by mutagenized method, and pilot study the culture condition of the strain .To sum up the anciently researchful fruit, the experimentation make some breakthrough at trehalose nature determination and quantify fixed through abundant tests and analysis. The experiment detect high trehalose content in the liquid-of yeast fermentation and find a new way for the continuity of the task.
