The condition of Aspergillus Oryzae 3042 producing Aminoacylase inductively was studied. 1%CSL is a good inductor in the condition of pH7.0, 26.5 0C, 1 60rpm/mm, 88hours yielding aminoacylase 53.1 i-i mol/hlmg, which was purified by precipitation of ammonium sulphate, SOURCETM 15Q&1 5S anion exchange chromatography. The purified protein showed a singleband on SDS-polyacrylamide gel electyophoresis with a molecular weight with a Mr. 7OkD and a specific activity of 3694.4 ~t mollhlmg. Of the seven acetylated amino acids tested, N-Ac-L-Tyr and N-Ac-L-Cys were the most favored substrates .The optimal pH and temperature for the enzymatic activity were 7.5 and 50 0C, respectively. It is activated by Co2~ and Zn2~ while inhibited by Ni2~ Cu2~.
