Sweet Tea as the main research raw material,determination conditions,extraction conditions by refluxing extraction and ultrasonic extraction,purification process and antioxidant activities of total Sweet Tea polyphenol were studied.The main experimental conclusions are as follows:(1)Using FC colorimetric method for determination of total Sweet Tea polyphenol,optimum conditions were as follows:2 m L FC reagent,7.5 m L 15% sodium carbonate solution,treatment at 30 ℃ for 60 min.The average recovery was 100.23% and the average relative deviation was 0.45%.FC colorimetric method can be used for content determination of the total Sweet Tea polyphenol.(2)Through dynamic and static experiment,HPD100 was optimized for purification of total Sweet Tea polyphenol from 10 different types of macroporous resin.The purification conditions of total Sweet Tea polyphenol by HPD100 were:concentration of sample 2.5 mg/m L ~5.0 mg/m L,PH = 4,the 2 BV/h adsorption velocity,150 m L 30% ethanol solution as eluent and the purity of sample purified was up to 63%.(3)Sweet Tea polyphenol sample got from HPD100 macroporous adsorption resin can be further purified by Sephadex LH-20 gel column chromatography.The purity of sample by 50% acetone elution was up to 70.3%.The experiment results of UV and HPLC showed that the sample contains gallic acid and catechins.(4)Antioxidant activities of Sweet Tea polyphenol samples before and after purification were studied.The results showed that the samples of Sweet Tea polyphenols before and after purification had higher antioxidant activities.
