Pine nut is two years of growth cycle, and contains rich nutrients, mainly contain fat, protein and vitamins. Pine nut oil mainly contains linoleic acid, alpha linolenic acid, oleic acid and various types of unsaturated fatty acid. It also contains sugars, minerals, amino acids the human body needs, and a variety of trace elements..In today’s society, hypertension is a common disease, leading to diseases such as stroke, heart disease, coronary heart disease(CHD). It has become the focus of the world’s health problems. Research suggests that angiotensin transferase(Agiotensin Converting Enzyme, ACE) inhibitory peptides in the renin angiotensin system, by reducing angiotensin transferase activity, and blood pressure to drop. In recent years, foodborne ACE inhibitory peptides has non-toxic side effects, high safety and low cost features, and become our research hot spot in the bioactive peptide of lower blood pressure.Albumin, globulin and glutelin obtained by Osborne method and Pine nuts protein isolate were investigated for their functional properties. The four proteins were determined to be 48.02%,81.93%,83.02% and 89.69%.To essential amino acids in content of total amino acid,albumin, globulin, glutelin and protein isolate values are 31.13%, 34.22%, 30.30% and 34.54%,respectively. Water holding capacity and oil holding capacity of glutelin are the highest and the vales are, respectively, 2.06mL/g and 3.98mL/g. To water holding capacity, the minimum value of is 0.59mL/g about protein isolate. To the value of oil holding capacity is the lowest and is 2.11mL/g. Globulin is the highest in protein solubility and the protein solubility displayed the lowest solubility were at pH 5.0. For exposed sulfhydryl content and total sulfhydryl content, the sequence from high to low is albumin, protein isolate, globulin and glutelin. To the highest of disulphide content is protein isolate about 28.74μmol/g and the lowest is glutelin about 13.46μmol/g. Foaming properties and emulsion properties of these proteins has the lowest values at pH5.0. Increasing with protein concentration, foaming properties and emulsion properties are also Increasing. SDS-PAGE showed that four proteins had different molecules.In this study, Pinus koraiensis Sieb.et Zucc protein isolate was hydrolyzed with alcalase to prepare antihypertensive peptides. Operating parameters such as pH, temperature, substrate concentration and enzyme dosage were optimized using response surface analysis based on degree of hydrolysis and ACE inhibitory rate. The optimum hydrolysis conditions for ACE inhibitory rate(70.1%) and degree of hydrolysis(17.5%) were found to be hydrolysis at 55 ℃, pH 8.9 for 3 h with an enzyme/substrate ratio of 7500U/g and substrate concentration of 2.5%, With the model prediction.Enzyme solution of the Alcalase pine nut protein is separate by ultrafiltration. Selecting two intercepting 3 kDa and 10 kDa membrane molecular weight, it can separate three components. Respectively investigate the ACE inhibition rate of three components, the results show that the A2 namely 3 kDa- 10 kDa. The ACE inhibitory is 74.8%. By Sephadex G-25 to purification of A2, under the condition of the gel filtration chromatography will be divided into three components, including B3 of the ACE inhibitory rate is up to 79.8%. For ACE inhibitory rate highest components of Sephadex G-15 further purification, it get the composition of the purified determination of ACE inhibition rate reached 93.1%. High reverse phase liquid phase separation and purification of C2 is a single refinement. Mass spectrum identification of pine nuts fall blood pressure of ACE inhibitory peptide molecular weight is 328.15 Da, according to the manual measuring molecular weight determining amino acid sequence structure may be Tyr- Phe.This study develops resource structure, properties about the pine nut protein of Changbai mountain and analysis of pine nuts fall blood pressure peptide. It can improve pine nut edible value and economic value. It also provide theoretical basis for further study on pine nut and scientific proof.
