Atherosclerosis is one of the most important factors which can cause cardiovascular, and the main cause of atherosclerosis is the high concentration of low-density lipoprotein(LDL). LDL apheresis is one of the most efficient methods to reduce LDL. LDL absorbent is the core part of LDL apheresis, and anionic absorbent has been most concerned because of its low price, extensive material selection and good absorption effect. It has been found that glutathione(GSH) has better binding effect with LDL than heparin and dextran sulfate, which are the most popular traditional anionic absorbents. Therefore, it is promising to invent a novel and efficient LDL absorbent with GSH as ligand. The main contribution of the thesis is described as below.(1) A novel PBT-GSH anionic absorbent is successfully prepared with PBT as the base and GSH as the ligand. GMA is chosen as the intermediate of GSH grafting PBT. In the first step PBT is modified with GMA, and then GSH is used to modify PBT-GMA in alkaline environment. Material weighing, ATR-FTIR spectroscopy, XPS elemental analysis and SEM microstructure observation are used to confirm the success of modification for PBT. The best conditions for GSH grafting on PBT-GMA has been analyzed. The result shows the best conditions for grafting is 80℃ for temperature, 24 h for reaction time and pH 12.5~13.0 for reaction environment.(2) Experiments are designed to detect the absorption property of PBTGSH by comparing with control experiments. After the separated parts of plasma centrifugation are weighed, it has been found that PBT-GSH can absorb more plasma protein which can be separated by centrifugation, and because LDL exists more in those proteins, it can be considered that the novel absorbent can selectively absorb LDL. After the total plasma protein concentration is measured by BCA method, it has been found PBT-GSH can absorb more plasma protein. After the LDL concentration is measured, it has been found PBT-GSH can selectively absorb LDL.
