With the rapid development of China’s aquaculture,fish processing industry has been developing rapidly in our country.A large number of leftovers will be produced in the process of fish,Such as fish head,bone and skin,scales and internal organs,etc.But they are mostly treated as waste away,and utilization rate is very low.The amount of scales account for about 1%- 5%of the amount of fish weight.If we can take advantage of them,not only can realize its high value,but also can promote the development of fish industry.At the same time,it also can reduce pollution of the environment and create a good economic and social benefits.This paper extracted collagen using the scales as raw material.Using collagen as the research object,we explore the best preparation technology of collagen peptides by single factor and orthogonal experiments.Through the establishment of lipid metabolic disturbance model,we research the influence of collagen peptide on the experimental animal weight,lipid metabolism,liver tissue and the antioxidant system.The main research contents and conclusions are as follows:( 1) Fish scales were decalcified method of cleaning,acid,crushing,boil adhesive and spray drying process to obtain high quality scales after collagen raw materials.In this paper,their physical and chemical properties of the scales collagen were tested.The test results show that the main ingredient in gelatin is protein.It’s content is up to 95%.As can be seen from the electrophoresis bands,it’s molecular weight distribution is in the tens of thousands to tens of Dalton.Glycine and Proline account for 50% of the total amino acids in scale collagen protein.(2)This article selects the alkaline protease in hydrolytic enzyme which is suitable for industrialized production,It explores the optimum process conditions of preparation of collagen active peptides.The study found that the substrate concentration has a little impact on the hydrolysis of collagen.We can choice of different substrate concentration according to the needs of production.The optimum technological conditions of enzymatic hydrolysis was enzyme solution temperature60 ℃,digestion time 6.0 h,pH value of 9.0,1.0% with enzyme.(3)The article explores the collagen active peptide in high-fat diet rats serum biochemical indicators and its the influence on liver pathological changes.Through the study we found that a high-fat diet can cause the symptoms of obesity and high cholesterol in rats,and all of a high-fat diet were produced the phenomenon of fatty liver in rats.At the same time we can see taht feeding collagen and collagen active peptide can improve the degree of liver fatty liver in rats,delay the fatty liver.It proves the collagen active peptide has the effect on the protection to of liver.(4)This paper designs a factory that the annual output of collagen active peptide product is 800 tons.This study does profound research of the main process,and select the related equipment and design the plane of the factory.In this paper,the design of collagen peptide powder factory Choose peptides in the production of advanced process equipment.It can realize automation, continuous,mechanized production, which saves resources, protect the environment.
