Study On Separation, Purification And Structure Of Polysaccharide From Scallop(Patinopecten Yessoensis)

Polysaccharide (SVP) was extracted from scallop viscera by papain. By a further separation and purification, we obtained a homogeneous polysaccharide (SVP-12), and the molecular weight, composition, primary structure of this polysaccharide were determined.SVP was treated with two procedures proteasea-Sevag method to deproteinate the polysaccharide. The reliquus protein was deleted by dialysising and freeze thawing. The scallop viscera polysaccharide was obtained by cryodesiccate, and its protein content was only21.0%.Then the crude SVP was purified by Sephacryl-S200gel and DEAE-cellulose52chromatography, finally we got the purified SVP-12fraction. It was a homogeneous polysaccharide demonstrated by HPLC, CAM and UV spectrum. Its molecular weight was about170kDa. Monosaccharide composition analysis indicated that SVP-12was composed of Rha, Fuc, Ala, Xyl, Man, Gla, Glc with molar ratio of1.65:2.54:4.05:5.60:1.48:4.90:1.00. It contained12.57%sulfate and8.46%aminohexose, aminogalactose was6.96%among the aminohexose.The structure of the SVP-12was investigated by using various spectroscopic techniques and chemical methods. The results of partial hydrolysis, Periodate oxidation, Smith degradation, Nuclear Magnetic Resonance and GC-MS dealing with methylation showed:(1) The main chain was composed of arabinose, fucose and galactose;(2) In the SVP-12, Ara was existed in the form of1â†',1â†'2,1â†'3, Fuc1â†',1â†'3,4, and Gal was existed in the form of1â†',1â†'4,1â†'6,1â†'3,4;(3) The main chain was Fuc, Ara and Gal repetitive branched structure, Fuc was existed in the form of (1â†'3,4) inoxidable glycosidic bond, Ara was existed in the form of (1â†'3) inoxidable glycosidic bond and was existed in the form of (1â†'3,4); Xyl (1â†'5) and Man (1â†'4)(1â†'2,4) were bisided the main chain.
