Realgar as traditional Chinese medicine plays an extremely important role in our country. Although realgar can treat many many diseases, but it belongs to a kind of arsenic-containing mineral drugs, which make it own certain side effects. Therefore, realgar has two characteristics of cure and pathogenic. And the toxicity of arsenic is closely related to its speciation. The study of realgar metabolites in animals is the basis to understand the mechanism of realgar’s cure and pathogenic.Organic arsenicals as feed additives can prevent and treat poultry coccidiosis, improve feed conversion rate, promote growth, improve the feather colour and increase egg production rate and pigmentation. Thus organic arsenicals have been widely used in livestock and poultry breeding in our country. But they have certain toxicity, excessive addition can do harm to the nervous system of animal, and may produce residual in animal body or break down into other toxic arsenic compounds.The achievement obtained in this paper as follows: (1) The method of HPLC-ICP-MS was established to analyse AsB, DMA, As(Ⅲ), MMA and As(V) in rat liver and kidney. Through optimizing the experimental conditions, Dionex IonPac AS19was chosen as analytical column,20mmol/L (NH4)2CO3was chosen as mobile phase, the pH and flow rate of the mobile phase were tested on the separation effect of five arsenic compounds. Ultrasonic water bath extraction was used to extract arsenic compounds in rat liver and kidney. Experimental results show that40Ar35Cl does not interfere with the determination results of each arsenic compounds, the linear range of five arsenic compounds is between0.5and300μg/L, the linear correlation coefficients (R2) are all greater than0.9990, the limits of detection are in the range of0.1to0.2μg/L, the standard addition recoveries of this method are from84.1to110.5%, and RSDs are less than5%. The study of total arsenic shows that total arsenic content in kidney is greater than the total arsenic content in the liver in the same concentration group, which indicates the ability of kidney to accumulate arsenic is greater than the ability of liver to accumulate arsenic. The results of arsenic speciation analysis showed that the main form of arsenic in rat liver is DMA after realgar exposed, a small amount of As (Ⅲ) and unknown compound2are also found in liver. The arsenic speciations are mainly DMA and MMA in rat kidney, containing a small amount of As (Ⅲ), unknown compound1,2and3.(2) The method of HPLC-ICP-MS was established to analyse arsanilic, roxarsone,4-hydroxyphenyl arsenic acid,4-nitrophenyl arsenic acid, DMA, As(Ⅲ), MMA, As(Ⅳ) in livestock and poultry meat products. With methanol and water (1:1, v/v) as extraction agent, ultrasonic water bath extraction was used to extract arsenic compounds in samples, Dionex IonPac ASM was chosen as analytical column,50mmol/L (NH4)2HPO4(pH=7.3) was chosen as mobile phase, then the arsenic compounds in livestock and poultry meat were determined. Experimental results show that the linear correlation coefficients (R2) of eight arsenic compounds were greater than0.9990in the range of1.0to300μg/L, the limits of detection are from0.1to0.5μg/L, the standard addition recoveries of this method are from80.8to105.7%, RSDs are less than5%. Through the analysis of real samples, arsanilic and roxarsone were detected in livestock and poultry meat, and one unknown arsenic compound was also check out.(3) The method of HPLC-ICP-MS was established to analyse arsanilic, roxarsone,4-hydroxyphenyl arsenic acid, AsB, DMA, MMA and inorganic arsenic in the pig concentrated feed. With methanol and water (1:1, v/v) as extraction agent, ultrasonic water bath extraction was used to extract arsenic compounds in samples, and oxidation treatment, Dionex IonPac ASH was chosen as analytical column,50mmol/L (NH4)2HPO4(pH=9.0) was chosen as mobile phase, then the arsenic compounds in pig concentrated feed were determined. Experimental results show that the linear correlation coefficients (R-) of seven arsenic compounds were greater than0.9990in the range of1.0to300μg/L, the detection limits are from0.1to0.5μg/L, the standard addition recoveries of this method are from79.9to112.2%, with precision examining method reproducibility, RSDs are less than5%. Through the analysis of real samples, AsB, DMA, iAs and p-ASA were detected in the pig concentrated feed.
