In the study, natural type II collagen was extracted from pig nasal bone which was the material. Collagen peptide was obtained from type II collagen by enzymatic process of Alkaline protease. Properties and anti-arthritic effects in mice of natural type II collagen and collagen peptide were investigated. The study had the vital significance for the anti-arthritis.Through the single factor experiment and response surface analysis, the optimum extraction process of type II collagen was obtained:NaCl of2.5mol/L, extraction time for30h, temperature at15℃. Under this condition, the extraction rate of pig nasal bone was22.58%. The purity was high up to82.86%. The results of determination of properties of collagen showed that the molecular weight was greater than300KD and the characteristic absorption peaked at230nm. This conformed to the properties of collagen. Collagen peptide was obtained from type II collagen by enzymatic process of alkaline protease. The molecular weight of collagen peptide was less than20KD. Results of HPLC-MS showed that there was no high molecular weighted protein fragments in collagen peptide.Effect of type II collagen and collagen peptide on collagen-induced arthritis model (CIA) was studied. By the anti-arthritic experiment in vivo, we concluded that the joint index of type II collagen group was2, and joint deformation rate was30%. The joint index of collagen peptide group was3, and joint deformation rate was70%. The cartilage degree of type II collagen group was low, and obvious degradation was appeared in collagen peptide group. Type II collagen had anti-arthritic function, but collagen peptide did not have.Type II collagen could protect immune organs by increasing thymus index (P<0.05) and reducing spleen index (P<0.05). The results of immune experiments showed that Type II collagen could improve the phagocytosis ability of macrophage (P<0.05) and improve proliferative ability of spleen cell (P<0.05). Type II collagen could protect and restore the ability of immune system. Result of detection experiments about IL-17, IL-10, MMP-3, TNF-a in serum showed that type II collagen could effectively reduce the level of IL-17(P<0.05), MMP-3(P<0.01) and TNF-a (P<0.05), and improve the level of IL-10(P<0.05). Type II collagen could reduce the extent of damage and oxidation of body. The collagen peptide didn’t have all these effects. Type II collagen had anti-arthritic function, but collagen peptide did not have.
