The optimal extraction conditions of persimmon tannin by using organicsolvent extraction method was inspectively achieved by single factor andorthogonal experiments. The optimal conditions for organic which would appeardryer "sticky wall" problem. The crude persimmon tannin extracted by organicsolvent extraction method were as follows: the extraction at80℃, the hydrochloricacid concentration of0.8%, solid-liquid ratio of1:10(w/v). Under this condition,the crude persimmon tannin yield calculated was19.8%, while the urified tanninseluent through rotary etotal phenols content of the crude tannin measured was28.56mg/g, and the content of condensed tannin was22.32mg/g.This article adopted the macroporous resin purification process to purify crudeextract of persimmon tannin. The purified tannins eluent through rotary evaporatorwas then innovatively freeze-dried to dried powder of a more ideal solid state,compared to vacuum drying tannins which would appear dryer "sticky wall"problem. The crude persimmon tannin extracted by organic solvent extractionmethod and the higher molecular weight persimmon tannin (HMWT) afterclassification were experimented by HPLC analysis and HPLC-ESI-MS analysis.Results showed that the higher molecular weight persimmon tannin containedEpigallocate-chin gallate (EGCG), epicatechin gallate (ECG),(Table)epigallocatechin (E) GC and another material which contained gallate substances.Meanwhile, clearing DPPH· abilitiy, reducibility and clearing the hydroxylradical scavenging capacity of persimmon tannin after the UF polymer grade weremeasured, results showed that HMWT has good antioxidant capacity and restorecapacity. Eventually, add different concentrations of HMWT to rice and lard oil,and then put it urified into electric oven substance to blast mandatory oxide, byantioxidant effect contrast of commonly used synthetic antioxidants such as BHT,TBHQ, the results showed that antioxidant capacity of the same concentrationHMWT was greater than BHT, slightly less than TBHQ. Tannins very difficult tosynthesize and is also easily soluble in water.The extracted antioxidant experimental validation not only provides the possibility for the persimmon tanninas natural antioxidants in foods application, but also provides direction for the deepprocessing of persimmon in China.
