Abstract:Ginkgo biloba contains rich pharmacological active ingredients. Many Research showed that Ginkgo Biloba Flavonoids had very high value in medical study. This article was about the extraction and separation of Ginkgo Flavonids. A new method utilizing hydrophilic ionic liquid aqueous two-phase system (ATPS) and hydrophobic ionic liquid liquid-liquid extraction was established. It provided a scientific basis for the development and application of Ginkgo Flavonids. The main contents and conclusion were as follows:Firstly, The binodal data of liquid-liquid equilibrium which was formulated by ionic liquid (IL)[Cnmim]Br(n=2、4、6、8),[C4mim]BF4,[Cnmim]N(CN)2(n=2、4、6、8) and salt (Na2SO4、(NH4)2SO4、NaH2PO4、 C6H5O7Na3) were determined. The factors influencing the phase-forming ability, IL structure, types of salts and the temperature of system were studied. The tie lines of four ILs [C4mim]BF4,[C4mim]N(CN)2,[C6mim]N(CN)2,[C8mim]N(CN)2ATPS were analyzed. The result was satisfactory when the binodal data were fitted by the equations.Secondly, In this paper, the phase-forming ability of three ILs [Cnmim]N(CN)2(n=4^6,8)and two salts (NH4)2SO4, NaH2PO4was studied. The factors affecting extraction efficiency, such as type of IL type and concentration of salt were discussed. Subsequntly effects of the system temperature, pH value, the amount of electrolyte (NaCl) on extraction efficiency had been investigated in optimal IL/salt ATPS.Thirdly,[C4mim]BF4/(NH4)2SO4ATPS was used to extract Ginkgo Flavonoids. The interaction of three factors like the amount of IL and salt, the system temperature was determined by central composite design (CCD) and response surface method (RSM).Lastly, Four type hydrophobic ILs [C4mim]PF6,[C6mim]PF6,[C6mim]BF4,[C8mim]BF4were selected as solvent to extract Ginkgo Flavonoids in the experimrnt. Effects of the structure of IL, IL concentration, temperature of the system, pH value, the amount of electrolyte (NaC1) on extraction efficiency had been investigated at the same time. Ginkgo Flavonoids were separated from IL-phase, and IL was recoveryed and recycled furthmore.In conclusion, when [Cgmim]BF4was selected as solvent to extract Ginkgo Flavonoids in the experiment, the extraction efficiency was higher than other ATPS.
