The components content of nitrogen and phosphorus are increase rapidly asalong as the promoting of industry, agriculture and human life level. Because of it, Itcauses the water eutrophication. In the water quality of eutrophication, theeutrophication of lake is the most serious. Because it leads to the environmentalproblems of world. It has been focused on people all over the world. The seriouswater quality of lake eutrophication can eventually lead to the "water bloom"phenomenon. The microcystin algae toxin (MCs) is a natural ring seven peptide livertoxins. The process of redox metabolism or broken make blue-green algae to secretetoxins. MC-LR has been known that is the strongest and the most common. Thehealth of human and animal have been affected severely by MC-LR.In the first chapter, In view of MC-LR physical and chemical properties, I gropegradually for setting related parameters that liquid chromatography electrospray massspectrometry technology method test the content of microcystin algae toxin in water.Through the research method of MC-LR, It systematically analyzes the molecularstructure and fracture mechanism of microcystin algal toxins. The result show that thestandard linear equation is Y=63.235756x-63.235756. The linear correlationcoefficients of MC-LR is0.997. The limits of quantification of MC-LR is0.01μg/L.The recovery is ranged from84.8%-95.3%. The relative standard deviation rangefrom2.7%to5.3%. The retention time of MC-LR is10.5min.In the second chapter, It introduces mainly the method of optimizing theexperiment research about extracting MC-LR. The first part of this chapter is that thereal samples have been collected. The real samples have been concentrated inprcessing. The real samples have been preserved in low temperature environment.The temperature has been setted at-20℃. The secondly through the experimentalcontrast, I select to add1.0mL of50%methanol as extraction agent. The extractiontemperature has been setted at40℃. The eventually through the principle of freezingand thawing. The algae solution of being condensed have been extractted toxins in theway of constant temperature and water bath ultrasonic. In the third chapter, It introduced mainly the influence study that theenvironmental factors impact on MC-LR of real samples. Through the method ofdetecting MC-LR standard samples, It detects the content of MC-LR. The samplecotent of June is the largest in six mouths. The sample cotent of June is less than thecotent of national standard. In the aspect of environmental factors, the analysis resultsshow that the relationship is a positive correlation between MC-LR and temperature,light, pH, nitrogen and phosphorus within certain limits. If the environment factorhave been changed, the content of MC-LR have been changed.In the fourth chapter, It introduced mainly the influence study that environmentalfactors impact on degrading MC-LR of real samples. Through the method ofdetecting MC-LR standard samples, It analyzes environmental factors impact ondegrading MC-LR of real samples with biological degradation. The analysis resultsshow that the relationship is a positive correlation between MC-LR and temperature,light,pH, excrept for nutrient.In summary, This text took MC-LR as object of study. Establish a method forMC-LR in water by liquid chromatography Electronic Spray Ion tandem massspectrometry. The method was rapid and accurate, easy to operate for MC-LR. Thisstudy provides the theoretical basis for analyzing molecular structure and internalfoim fracture mechanism of MC-LR. It also provides technological support that theenvironmental factors impact on production and degradation of MC-LR.
