To demonstrate the temperature of Saccharomyces cerevisiae cell wall proteome affected by DTT dissolved combining phenol extraction method to extract the cell wall proteome, and the use of two-dimensional electrophoresis and mass spectrometry to analyze them. The results show that the 2-DE map of detected protein spots 28 ℃ for 539, 37 ℃ to 540. Which has a protein spots disappeared on 37 ℃ map, two protein spots were up-regulated the expression, the expression of two protein spots down, two protein spots on the map appeared only 37 ℃, the use of mass spectrometry were identified six different proteins. These differences indicate improved protein cultivation temperature had affected expression of certain proteins on and off specific genes within the cell wall, high temperature stress Saccharomyces cerevisiae FFC2146 cell wall proteins and small molecules Ssa2 new guanosine triphosphatase, inorganic pyrophosphatase upregulated, while the lack of pyruvate kinase disappear, while the glucose-6-phosphate dehydrogenase, and Glyceraldehyde-3-phosphate dehydrogenase expression down. These results indicated that the heat shock protein Ssa2 protect intact cell wall at a high temperature, the cells continued growth and reproduction; EMP glycolytic pathway of Saccharomyces cerevisiae under heat stress is blocked, in the role of transketolase glycolytic pathway turned HMP pathway, Get enough energy to maintain normal cell metabolism.
