The importance of Magnetic Resonance Spectroscopy (MRS) in measuring metabolite concentrations in vivo is increasing by the time. And it is a hot topic that quantitative measuring GABA concentrations by MRS in vivo. However, it is difficult and faces various challenges from the lower concentrations and the overlapping of peaks from some other metabolites. So the traditional measuring methods could not be used in separating GABA and quantitative measuring it.In this paper, we separated GABA from other overlapping metabolites and made a quatitative measurement of GABA concentrations in vivo. Firstly, based on the principles of MR, some theory and methods of analyzing and design pulse sequence were introduced. After that, we studied the classical Bloch equation. With it, we analysed how to export the free induction decay (FID) signal in the single pulse sequence. Secondly, the principle of MR scanner and performances of some hardware were studied for the future programming work. Finally, based the principle of MRS, the principles of spectral analyze and algorithms of quantitative measurement were studied with the help of some software packages, with which the data from phantom and brain was processed and GABA was separated.To point out, the methods of measuring GABA concentration with special pulse sequence and software package can be used to measure the other metabolites with lower concentration in brain in vivo, which will make progress in the study of brain, physiology and pathology of functional disturbance and development of new medicines.
